阿司匹林和塞来昔布对乳腺癌细胞MCF-7增殖影响的实验室研究

【摘要】 目的:通过研究非甾体抗炎药物( Nonsteroidalantiinflammatory drugs, NSAIDs)阿司匹林和塞来昔布及二者分别联合阿那曲唑对人乳腺癌细胞MCF-7增殖的影响,探讨并比较阿司匹林和塞来昔布的抗肿瘤作用。方法:采用MTT法检测阿司匹林和塞来昔布及二者分别联合阿那曲唑对乳腺癌细胞MCF-7增殖的影响。结果:通过MTT法检测抑制率的结果得出阿司匹林和塞来昔布对MCF-7细胞生长有抑制作用,且药物剂量越大,作用时间越长,抑制作用越明显,差异具有显著性(P<0.05);同时亦得出阿霉素对MCF-7的抑制作用亦呈剂量﹑时间依赖性,差异具有显著性(P<0.05)。此外,阿那曲唑和阿司匹林或塞来昔布联合后对MCF-7细胞的抑制作用较单用阿那曲唑增强,且剂量越大,抑制作用越明显,差别具有显著性(P<0.05)。结论:阿司匹林和塞来昔布均有抑制乳腺癌MCF-7细胞的作用,并且呈时间、剂量依赖性;二者与阿霉素可达到几乎相同的抑制乳腺癌细胞MCF-7增殖的作用;阿司匹林、塞来昔布分别与阿那曲唑联合应用可起到协同抗肿瘤的作用,呈剂量依赖性,差别具有显著性(P<0.05)。
【Abstract】 Breast cancer is a frequent tumor among women,incidence rate and mortality rate of that have beenincreasing for last twenty years. Breast cancer has becomethe most frequent cause of death among women. Recently, alarge of studies discovered that COX-2 over-expressed incolon carcinoma, gastric carcinoma, esophageal carcinoma,pancreatic carcinoma, lung carcinoma, breast cancer and soon commonly, that suggested COX-2 probably participated indevelopment of many tumors. NSAIDs is a antipyreticanalgesic, because it has good therapeutic effect thatrelieving fever, relieving pain, anti-inflammatory, andrelieving rheumatism, it has been one of the most commonlyused drugs. Nearly, we discovered that NSAIDs has theantineoplastic effect besides relieving fever andparegoric efficacy unexpectedly. Above all, aspirin caninhibit COX-1 and COX-2 simultaneously, and decreaseincidence rate of breast cancer. But the side effect ofnon-selective COX-2 inhibitor if long-term administration,especially the effect on digestive tract that confines itsapplication in preventing breast cancer. A selective COX-2inhibitor, celecoxib can prevent and treat colon cancer,that has been confirmed, FDA gave approval that celecoxibcould be used to treat familial polyposis on 2000, and thatcelecoxib was used to treat colon tumor had enteredclinical trial. At present, findings manifest selectiveCOX-2 inhibitors have preventive and therapeutical effecton breast cancer, furthermore they can cut down distantplace metastasis. Now, COX-2 inhibitors are used toclinical practice gradually, and they usually combine withother medicines. Initial researches indicated thatselective COX-2 inhibitors had preventive and therapeuticeffect on mammary cancer, they tolerance was better. Inthis study, the inhibitory effect that NSAIDs and arimedexon breast cancer cell MCF-7 was detected by MTT assay. Fromthe results, we comprehend that the effects of NSAIDs andarimedex on the growth of MCF-7.In this study, we adopted MTT assay to detect theseeffects of 2.5mM, 5.0mM, 10.0mM aspirin and 30uM, 60uM,120uM celecoxib on the growth of human breast cancer cellMCF-7.The results showed NSAIDs can inhibit the growth ofMCF-7, and these inhibitory effects were time-dependent anddose-dependent. The most inhibition ratio of aspirin andcelecoxib were 68.88% and 87% respectively, the inhibitionratio of 2.0uM ADM on MCF-7 for 72 hour was 86.3%. Theseresults that anastrozole combining aspirin and celecoxibrespectively affected on MCF-7 showed NSAIDs combiningwith anastrozole had synergism antineoplastic effect, andthese effects were dose-dependent.From these results, we concluded that both aspirin andcelecoxib had inhibition effects on the growth of humanbreast cancer cell MCF-7,and they were time-dependent anddose-dependent inhibitors. The two inhibitors can achieveidentical inhibition effect on MCF-7 compared to ADM.Aspirin and celecoxib combined with anastrozole havesynergism antineoplastic effect, which weredose-dependent. This study contributes to the method thataspirin and celecoxib could be used to prevent and treatbreast cancer.
