六种中药成分对细菌作用的微量量热法研究

【摘要】 中药是我国传统医学的重要组成部分,是治疗疾病的重要武器,已在医疗中应用了几千年。随着现代分离技术的发展,药物化学已成为化学、生物学科学者关注的热点。用物理化学的方法和热动力学原理来研究中草药活性成分对细菌代谢作用的影响是化学研究的一个重要领域。系统的研究中药有效成分的抑菌活性,进一步开发研究新药物,成为越来越多的科学家关注的目标。 本文应用微量量热法研究了五种中药及其有效成分对细菌生长代谢作用的影响,借助八通道微量量热仪对细菌正常代谢及中药成分对细菌生长代谢过程的热功率—时间曲线进行了测定,利用细菌生长的理论模型,进行热动力学性质的研究,其研究为新药的合成及特效药物的筛选提供定量的理论依据 对于中药有效成分的研究多集中于生物碱、强心甙、有机酸、氨基酸等,但用微量量热法研究中草药成分中黄酮、香豆素类对细菌生长代谢抑制作用的报道甚少。 主要研究内容 1.药物成分的提取 根据不同中药成分的性质,采用不同的实验方法对中药(北豆根、黄芩、蛇床子、虎杖、益母草)的活性成分进行提取。 2.中药成分对细菌生长代谢抑制规律的研究 为了确定药物对细菌生长代谢作用的规律和最佳用药浓度,分别测定了在不同浓度的药液作用下细菌生长代谢的热功率—时间曲线,用Logistic方程对其指数生长期的曲线进行处理,得到细菌在不同浓度药液中的生长速率常数,进而拟合出生长速率常数与药物浓度的关系,确定了最小抑菌浓度。 研究结果表明六种中药的有效成分对大肠杆菌的生长代谢都有抑制作用,但是抑制的效果不同,其中虎杖大黄素的抑菌效果最好,最小抑菌浓度为0.01621 mg/mL;益母草黄酮药液对金黄色葡萄球菌的生长代谢也具有抑制作用,其最小抑菌浓度为1.159mg/mL。

【Abstract】 The Chinese natural drugs play an important role in traditional medicine, which are one of the important weapons to cure diseases. They have been applied to the medical treatment for thousands of years. With the development of modern separation technology, the medicinal chemistry has become the hot spot of many scholars. Studying the influence of the active component of the Chinese medicine on the bacteria metabolism with the physical chemistry method as well as the thermodynamic theory is an important field for the chemistry research. The study of the bacteriostasic activity of the effective medicine components and further developing new drugs attract more and more scientists’s attention.The microcalorimetric method was used to study the activity of five kinds of drugs including their effective components on the bacteria metabolism, carries on the determination with the TAM Air to the bacterium normal metabolism and medicine solutions to the bacterium growth metabolism process thermal power-time curve, the thermodynamic characters were studied by using the theoretical model of bacterium growth, these may provide the theory basis for the new medicine synthesis and medicine screening.Many researches on the effective components of the Chinese medicine are about the alkaloid, the strong heart glucoside, the organic acid, the amino acid and so on. Studying the effect of the flavone, coumarin on the bacterium growth metabolism by microcalorimetry is really few. 1.The chief content 1.1 The extraction of drugsDepending on the nature of Chinese medicines, the effective components of drugs were extracted in suitable ways. 1. 2 The study of the regulation of the bacteria growing and metabolizingin culture medium containing drugs at different concentrationsIn order to find the regulation of the bacteria metabolism and the optimum concentration of drugs, drugs of different concentrations were put into the liquidmedium containing bacteria to get suspending liquid. The power-time curves of bacterium at 31 OK were analyzed using logistic equation so that the growth rate constant of bacterium in drugs of different concentrations was obtained. Then the relationship between growth rate constant and concentration was determined and the optimum concentration was confirmed.The findings indicated six kinds of medicine solutions have the inhibitory action to the E. coli growth metabolism, but the bacteriostasis effect is different, giant knotweed emodin have the best bacteriostasis effect and the smallest bacteriostasis density is 0.01621 mg/mL;Leonurus flavoe to the S.aureus growth metabolic also have effect, the minimum bacteriostasis concentration is 1.159mg/mL.Microcalorimetric method is a new method for studying the Chinese traditional drugs to bacteria metabolism. It can directly measure the thermodynamic process of system, and it needn’t add other reagents, so it will not introduce impurity, and not disturb natural metabolic process;It can not only afford thermodynamic data but also kinetic data.
