应用生物芯片技术对不同人群血清细胞因子临床意义的研究

【摘要】 细胞因子网络的复杂性及其任务体现在它对正常或病理状态的评价。因此,目前各实验室对测定同一样品的多种不同细胞因子的研究越来越感兴趣。对多种细胞因子的测定可以更好的用于临床对疾病的诊断及治疗。本课题应用生物芯片技术对不同人群血清12项细胞因子进行了测定研究,发现细胞因子在不同人群和不同疾病中均起着重要的作用。 一、因子芯片稳定性评价及其正常参考范围的建立:英国RANDOx公司生产的EVIDENCE180全自动化学发光生物芯片分析系统其细胞因子模板的每一张芯片可同时定量测定IL-1α、IL-1β、IL-2、IL-4、IL-6、IL-8、IL-10、VEGF、IFNγ、EGF、MCP-1以及TNFα12种不同的细胞因子项目,经稳定性研究证明其性能较好。细胞因子在健康人群中稳定存在,与年龄、性别无明显相关,作为评价机体免疫功能的一组指标,具有潜在应用价值。 二、在对SARS患者血清细胞因子的研究中发现,SARS患者血清中的细胞因子水平除IL-1β没有显著变化外,其他各项变化均比较明显。其中,IL-6、IL-8、IL-10及IFNγ与健康人比较显著上升;而IL-1α、IL-2、IL-4、VEGF、EGF、MCP-1及TNFα与健康人比较显著下降。各种血清细胞因子·在SARS患者不同发病时期有较大变化,发病旱期尤为明显,本结果进一步支持SARS患者的病理损伤过程与免疫失调有关。 三、青少年女性中长跑运动员血清EGF显著低于正常人,5周大运动量集训后血清IL-8、VEGF、TG、CH、HDL、ALB、GLU、Na、K和CL显著下降,EGF、Ua显著上升。纳米力谷源胶囊对运动应激后血清IL-1β、IL-2、EGF、TG、GLU的调节作用较为显著。因此,大运动量的集训可导致运动员机体应激性损伤,免疫功能紊乱,并进而导致机体—系列生理生化的变化。纯中药制剂——纳米力谷源胶囊可有效的增强运动员机体的应激能力,提高运动员的免疫力和抗疲劳能力。 四、服用FK506的肾移植患者,其血清中IL-6、IL-8、IL-10比正常人
【Abstract】 Knowledge of the complexity of the cytokine network and the role played by cytokines is critical in understanding normal and pathological processes. Therefore assaying a number of cytokines in the one sample has become of increasing interest in laboratory medicine. The knowledge gained from multiple cytokine analysis should allow better diagnosis and disease management. In this study we detected 12 serum cytokines of various people group by using biochip technology, and the results showed us that cytokines played a very important role in human body.1. The EVIDENCE 180 analyser is a fully automated biochip array system. The cytokine panel can quantitatively test for IL-1α , IL-1β , IL-2, IL-4, IL-6, IL-8, IL-10, VEGF, IFN γ , EGF, MCP-1 and TNF α simultaneously. Serum cytokines is very stable on healthy population and it is no correlations with ages and gender. It is very useful to evaluate the functions of body’s immune system.2. The dynamic studies on the serum cytokines level of different stage in the diagnosed SARS patients. Compared with healthy group, IL-6 、 IL-8 、 IL-10 、 IFN γ increased obviously and IL-1 α、 IL-2、 IL-4、 VEGF、 EGF、 MCP-1、 TNF a decreased obviously. Whereas IL-1β has no statistic changes among different stages of SARS. There were obvious changes of multiple cytokines in different phase of SARS pathological process, especially in the early phase. It is further support the hypothesis that over-reaction of the immune system initiated the pathological injuries of the patients.3. Compared with healthy population EGF is lower obviously. After heavy intensity training of 5 weeks IL-8、 VEGF、 TG、 CH、 HDL、 ALB、 GLU、 Na、K and CL decreased obviously and EGF > Ua increased obviously. Nanometer-particulate LiGuYuan capsule can influence serum IL-1 P > IL^ EGF> TG^ GLU evidently. Heavy intensity training caused immune ability decreased and nanometer-particulate LiGuYuan capsule can anti-fatigue and increase the capability of body’s respond.4. In the patients who after kidney transplantation and treated by FK506, the serum IL-6, IL-8 and IL-10 increased obviously, whereas serum EGF decreased markedly. The immune function of the patients after kidney transplantation was still abnormal and serum cytokines were very useful to follow up and monitoring the rejection of the transplanted organ in them.5. The dynamic studies on the variation of serum cytokines and biochemical parameters in post kidney transplanted patients, we found serum IL-6 and IL-8 had reached the highest level at 7 days after kidney transplantation, whereas IL-2 and IL-10 were reached the highest level at 14 days. Then they decreased gradually. The decreased VEGF, EGF and MCP-1 in kidney failure patient had increased after transplantation. UN, Cr, Ua and Glu had been restored to normal level after transplantation and there was obviously relationship between IL-8 and serum LDH. The variation of serum cytokines were related with the recovery of kidney function and tissue injury, it is a potential useful marker to follow up organ transplantation patients.6. In the stable stage of COPD, the level of serum IL-8 is increased and it is possible involved the processes of chronic airway inflammation.Based on these studies, there were very important significance of multi-cytokines in various patients, especially in the patients with inflammation disease and immune-disorder. So that, to detect various serum cytokines associated is very useful to diagnose and monitor the multi-disease.
