柴胡皂苷分离纯化研究

【摘要】 柴胡是我国传统中药中重要和常用的药材,具有显著的生理活性。其中柴胡皂苷是柴胡中主要的有效成分。研究发现柴胡皂苷具有解表和里、疏肝解郁、升举阳气之功效。因此一直备受药物研究人员的普遍重视,具有一定的开发前景。本文研究了柴胡皂苷的分离纯化工艺。以药用正品北柴胡为原料,经过大孔吸附树脂精制和硅胶柱层析分离纯化,得到纯度大于95%的柴胡皂苷单体。本文采用AB-8大孔吸附树脂对药材提取液进行了精制,以不同浓度的乙醇溶液进行梯度洗脱,再经萃取、脱色、结晶过程得到精制的柴胡粗皂苷,柴胡皂苷的含量有很大的提高,为后续工艺研究提供原料。以精制的粗皂苷为原料,调节不同展开剂的极性,通过硅胶柱层析的方法,得到了纯度在95%以上的柴胡皂苷单体6种。以柴胡皂苷a为检测指标,对柴胡提取工艺做了正交实验优化。实验发现了柴胡提取的最佳提取条件。

【Abstract】 Bupleurum is a very important and useful medicinal materials in traditional Chinese medicine, and its saikosaponins have very prominent physiological activity . So saikosaponins is the main active components in the bupleurum. It is regarded highly by the researcher and has a better tomorrow. This paper was concerned about the isolation and purification of saikosaponins from Bupleurum Chinense DC. It was refined by the macroporous adsorption resins and isolated by the silica gel chromatography. The purity of saikosaponin was above 95%. Firstly, saikosaponin was prerefined by macroporous adsorption resins, and the gradient elution was performed with ethanol. Then it was extracted, decolored and crystaled. And we can get finer bupleurum saikosaponins. At the end, using the refined saikosaponins as material, we got six kinds of monomer of saikosaponins by silica gel chromatography. Making bupleurum saikosaponin a as examine target, we optimized the extraction condition of medicinal materials by orthogonal design. And we found the best extraction condition.
