色素亲和色谱和流通色谱模型研究

【摘要】 色谱过程理论的研究对于色谱过程操作方法的设计和优化有着重要的意义。然而,其在多组分色谱和双孔介质填充色谱理论研究方面尚不完善。围绕色素亲和色谱理论和流通色谱理论,本研究展开了以下的研究工作:建立了色素亲和色谱的线性洗脱模型,该模型基于轴向扩散理论和孔扩散模型,吸附等温线采用双组分空间质量作用吸附平衡模型。本文研究了牛血清白蛋白和牛血红蛋白在Cibacron blue 3GA修饰的Sepharose CL-6B填充柱的在盐浓度梯度下的动态吸附行为。该模型可以很好的预测不同条件下的色谱洗脱曲线,说明双组分空间质量作用模型和色素亲和色谱的线性洗脱模型可以有效的描述蛋白质在非线性色谱中的传质行为。建立了流通色谱过程模型,称为堆积微管模型。模型假设双分散吸附剂粒子是由堆积在一起的微管组成的。为了验证模型,合成了双孔型,小孔型和大孔型三种阴离子交树脂,并对其参数做了表征。使用堆积微管模型对三种介质填充柱的穿透曲线做了模拟,堆积微管模型可以很好的预测不同条件下的三种色谱介质填充柱的洗脱曲线。相关系数验证结果表明,在所有的试验条件下使用堆积微管模型的模拟结果均比双线性驱动力模型的预测结果好,这说明对于使用致孔剂致孔的色谱介质来说,堆积微管假设比现有其他数学模型中的堆积微球结构假设更加合理。
【Abstract】 The mathematical modeling of chromatography is of particular interest tobioengineers, since it can be used as a useful guide and powerful tool in the optimaldesign of the operation scheme or scale up of a chromatography process. The followingwork has been done about theories for dye-ligand affinity chromatography andflowthrough chromatography:A mathematical model for dye-ligand linear-gradient chromatography of a binaryprotein system was established based on the axial dispersion theory and pore diffusionmodel. Bovine serum albumin (BSA) and hemoglobin (Hb) were used as model proteinsand Cibacron blue 3GA (CB) modified Sepharose CL-6B (CB-Sepharose) was used asadsorbent. The binary adsorption isotherms of the proteins were expressed by the stericmass-action (SMA) formulism. It was found that the model predictions agreed reasonablywell with the experimental data obtained under various conditions. It indicates that thebinary SMA formulism was useful for simulation of nonlinear chromatography.A mathematical model for flowthrough (perfusion) chromatography, namelypacked-microtubes (PMT) model, has been proposed for a column packed with biporousanion exchanger in which the micropores and flowthrough pores are created with liquidand solid porogens, respectively. The model is established based on the assumption thatthe biporous particle is made up of packed microtubes. Three kinds of anion exchangers(i.e., microporous, macroporous and biporous resins) are prepared and were used asadsorbents to determine the model parameters and to evaluate the model. It is found thatthe model predictions agree reasonably well with the experimental data obtained undervarious conditions, which suggests that the structure assumed in this model is reasonable.
