中药提取液絮凝流体动力学的研究

【摘要】 用絮凝澄清技术处理中药提取液时,不仅可以大大缩短中药液的生产周期,有效成分的保留率高,而且还可以提高制剂的稳定性和澄清度。本文即是采用絮凝法代替传统的费时耗能的醇沉法应用于乐仁堂养阴清肺糖浆产品的澄清过程,以达到缩短生产周期,降低生产成本的目的。本文选取了7种无毒无害的天然有机高分子物质为絮凝剂,分析了养阴清肺糖浆的水提液与醇提脱醇液的基本特性,采用烧杯混凝实验方法分别从上清液的剩余浊度和有效成分保留率两方面综合考虑甄选出了适合水提液与醇提脱醇液的絮凝剂并确定了其最佳的絮凝工艺条件。实验结果发现用ZTC1+1絮凝剂处理水提液时最佳工艺条件下可使药液的剩余浊度达到13.7NTU,甘草酸的保留率在83.2%以上; 用中药醇沉壳聚糖处理醇提脱醇液时在最佳工艺条件下可使药液的剩余浊度达到11.4NTU,芍药苷的保留率在94%以上。用高速离心法对絮凝后的药液进行后续处理,发现进一步提高了药液的澄清度和稳定性,高速离心法可以作为絮凝澄清技术的有效后续处理方法。从絮体尺寸、形态及其沉降特性分析了流体力学对絮凝过程的影响,搅拌强度与时间对药液的有效成分的保留率无太大影响,但对上清液的剩余浊度和絮体的沉降特性有一定的影响。本文还通过絮凝反应器的放大实验,考察了径向流与轴径向混合流两种不同形式的流场对絮凝效果的影响,发现径向流产生的较强的水流剪切力与较弱的全场混合程度不利于絮凝过程的进行,而轴径混合流强化了絮凝反应并缩短了反应时间。最后根据流体动力学对絮凝过程的影响分析为指导设计了格栅式的絮凝反应器,并由实验结果验证了其能够达到良好的絮凝效果。

【Abstract】 Compared with alcohol-precipitation method in clarification process of Chinese medicine extraction, flocculation can reduce the production cycle, lower the energy consumption, ensure the retention rate of available components as well as increase the clarity and stability of the pharmaceutical product. In this paper, by the way of flocculation substituting alcohol-precipitation, the Yangyingqingfei syrup produced by Lerentang is studied as a case. 7 kinds of natural macromolecule are selected as flocculants to treat Chinese medicine extraction. The jar test results show that, ZTC1+1 and chitosan can act as flocculants respectively in the process of clarification for water-extraction and alcohol-extraction. When under their optimum conditions in the process of flocculation for water-extraction and alcohol-extraction, the residue turbidity can reduce to 13.7NTU and 11.4NTU respectively, and the retention rate of avialible component can keep at 83.2%(glycyrrihizic acid) and 94%( paeonia radix). An experiment using centrifugal separation method as subsequent process after flocculation is carried out and the results indicate this method can further improve the clarity and stability of the pharmaceutical product with no effect on the available component of pharmaceutical product. Based on the analysis on flocs’ dimention, morphosis and setting properties, flocculation mechanism including the effect of hydrodynamics on flocculation was also studied systematically. Stir velocity and time show weak influence to the retention rate of avialible component but strong influence to residue turbidity and setting properties of flocs. Through the test of amplifying flocculator, it is found that axial-radial flow can reinforce flocculation process compared with radial flow. Under the direction of theory and above experiments, a kind of new type gridding flocculator is designed and further verified its flocculation effect by experiment. The experiment results show that, under identical average energy dissipation, gridding reactor has better flocculation efficiency than that of no-gridding reactor in clarification process of Chinese medicine extraction.
