应用圆二色光谱研究电场对蛋白质(酶)构象的影响

【摘要】 电场作为一种物理因素,对生物体的影响已成为生物物理的重要研究内容。特别是电场对植物种子的宏观生物效应及应用已有大量的研究成果。但宏观生物效应的微观机理是迫切需要解决的问题。它的研究,对于推动电场生物效应的深入研究,加快开发利用速度具有非常重要的意义。 本文采用圆二色光谱法测定α-淀粉酶、脂肪酶的圆二色图谱,研究电场对生物大分子构象的影响,特别是对二级结构的影响,为阐明生物大分子结构与生物体的功能之间的关系,揭示电场对生物体的作用机理奠定实验基础与理论支持。 研究结果表明:经不同强度的电场处理一定时间,α-淀粉酶、脂肪酶的二级结构的含量发生了不同程度的改变。其变化呈非单调型(振荡)关系,且种类不同变化趋势、程度也不完全相同。α-淀粉酶在电场强度为2.0kV/cm、4.0kV/cm时二级结构变化明显,脂肪酶在电场强度为1.5kV/cm、2.0kV/cm和4.5kV/cm时二级结构变化明显。 根据分子生物学理论和我们的实验结果,我们认为,电场处理种子改变了酶蛋白的构象,而且,不同电场强度对酶蛋白分子的构象产生不同影响,因此产生不同的宏观生物学效应。 但对于电场处理条件(处理时间,电场频率)对蛋白质(酶)二级结构的
【Abstract】 The effect of electric field, a kind of physical factor, on organism has already become important research contents in biological physics. It has already been carried out that a large amount of research about macroscopical biological effect on plant seeds and its application.The mechanism of the macroscopical biological effect is a problem to be urgently solved. And, the result of these studies is of great important meaning to scientific research of the electric biological effect and to promoted developing utilization.In this paper, We aim to study the effect of different intensity of the electric field on biological macromolecules structure, especially the secondary structure —using the spectrum of α-amylase and lipase treated with electric field of different intensity measured by Circular Dichroism spectroscopy, which is helpful to establish the experiment foundation and the theory for illustrating the relationship between the structure of biological macromolecules and the function of the organismand also to give the mechanism of the effect of electric field on the organism.The result of study show that the content of secondary structure has changed in various degree after α-amylase and lipase treated with the different intensity of electric field for fixed time , also the tendency and level of change are distinct from.a different kind. When the intensity of electric field is 2. 0kV/cm and 4. 0kV/cm, the change of α-amylase secondary structure is the most obvious and outstanding. However, to the lipase, it is in 1. 5kV/cm, 2. OkV/cm and 4. 5kV/cm.According to the theory of molecular biology and our experimental result, we think that the structure of the enzyme of plant seeds has been changed by the electric field and the different intensity of electric field has a different effect on the molecule structure of enzyme protein. Accordingly, this change will produce different macroscopical biological effects.The influence of treatment condition, including treatment time and electric field frequency on the secondary structure of protein and the relation between the conditions and macroscopical biological effect are pending for further discussion.
