氡及其子体和苯对小鼠血液和骨髓联合毒作用

【摘要】 目的:研究氡及其子体和苯对小鼠血液和脂质过氧化以及骨髓毒性的联合作用及其类型,为进一步探讨二者联合毒作用的机理提供实验依据。 方法:以BALB/c小鼠为研究对象,采用动式染毒法。根据3×3析因设计将小鼠随机分为9组,每组6只。氡及其子体的累积剂量分别为55、101WLM;苯的浓度为100、300PPM,每天5小时,每周5天,共染毒2周。以外周血细胞计数反映对血细胞的毒性。以超氧化物岐化酶(SOD)活性和丙二醛(MDA)的含量反映脂质过氧化程度。以骨髓粒细胞(CFU—GM)体外培养反映对粒系祖细胞的影响,用3H-TdR掺入法观察小鼠骨髓细胞DNA合成的改变。对实验结果进行析因分析,联合作用的判断依据单向法即E1+2为联合作用,E1为单独作用,如果E1+2>E1,则为增强作用。 结果:在实验剂量范围内,染氡高剂量组白细胞、淋巴细胞、红细胸计数显著性降低。染氡低、高剂量组外周血SOD活性显著降低,MDA含量的显著增加。染氡低剂量组CFU-GM集落计数、H3-TdR掺入显著性增加,高剂量组则显著降高。骨髓有核细胞计数无明显改变。 染苯高剂量组血小板、血红蛋白 显著性降低。染苯低、高剂量组白细胞、淋巴细胞、红细胞、SOD活力显著下降,而MDA含量显著增加。对CFU-GM、H3-TdR掺入无明显改变,但骨髓有核细胞计数显著性降低。 联合染毒组外周血细胞计数、血红蛋白含量均显著性降低,SOD活性显著性降低,MDA含量显著性增加。CFU-GM、骨髓有核细胞计数也显著性降低。析因分析结果表明:氡及其子体在H3-TdR掺入、CFU-GM集落计数、SOD、MDA等方面存在交互作用。 结论:氡及其子体使外周血红细胞、淋巴细胞数量减少,脂质过氧化增强,其对骨髓细胞的效应为双向性,即在低剂量时刺激骨髓细胞的增生,而高剂量
【Abstract】 Objective: To study the joint effects of radon and benzene on the blood and bone marrow of mice to provide experimental data for further study.Methods: The mice were divided into 9 groups by 3x3 factorial experimental design, with 6 mice each group. The accumulative doses of radon and its progeny were 55 and 101 Working Level Month (WLM). The mice were exposed to inhaled benzene (100 or 300 PPM) 5h/day and 5days/week for 2 weeks. Peripheral blood cells were counted to assess hematotoxicity. Superoxide Dismutase(SOD) and Malondialdehyde (MDA) were measured to show the degree of lipid peroxidation. CFU-GM reflected the effects of inhaled radon and benzene on haematopoieti progenitor. ~3H-TdR incorporation was used to observe the effect on regeneration of the bone marrow cells.Result: It was shown that the white blood cells (WBC), Lymphocyte(Ly ) and red blood cells (RBC) decreased significantly in the high dosegroup ,SOD decreased and MDA increased significantly in the low and high dose groups of inhaled radon and its progeny.Platelet (Plt) and Hemoglobin (Hgb) decreased significantly in the high dose group of inhaled benzene. WBC, Ly, RBC and SOD decreased and MDA increased significantly in the low and high dose groups of benzene exposure. CFU-GM and ~3H-TdR incorporation did not change obviously but BMC decreased significantly.In the combined groups of inhaled radon and benzene, peripheralblood counts, Hgb and SOD decreased significantly, but MDA increased significantly. CFU-GM and BMC also decreased significantly. It was shown that radon and benzene could have interactive effects on CFU-GM, 3H-TdR incorporation, SOD and MDA.Conclusion: Radon and its progeny can stimulate regeneration of the bone marrow cells at the low dose, but can depress the bone marrow cells at the high dose. The radon and benzene could bring out strengthened toxic effects in a joint manner.
