局灶性脑缺血再灌注大鼠CGRP、AchR及存活素的表达

【摘要】 [目的] 研究脑缺血再灌注后CGRP、AchR及存活素表达的变化规律,探讨CGRP、AchR及存活素在脑缺血损伤时的作用。 [方法] 成年健康雌性Wistar大鼠36只,应用线栓法建立大鼠大脑中动脉阻塞(MCAO)再灌注模型。运用HE染色及免疫组织化学等方法研究了脑缺血再灌注损伤时CGRP、AchR及存活素表达的时程变化。 [结果] HE染色及焦油紫染色显示,低倍镜下可见缺血中心区神经细胞坏死,缺血半影区神经细胞的密度减少,神经元缺血样变性,胞体缩小成三角形、扇形或长条形,细胞核固缩深染、核仁不清,胞浆呈空泡样。皮质神经元表现为胞浆疏松、核轻度变形、深染。变性神经元与正常神经元共存。再灌注14天后缺血中心区和半影区可见大量胶质细胞增生。脑缺血再灌注大鼠缺血侧大脑皮质再灌注后存活素的表达与假手术组比较明显增加,在6h内达高峰(P<0.001),3天时降至基础水平,但7天时有所升高(P<0.01),14天重新降至基础水平。而大脑纹状体脑缺血1h再灌流2h开始存活素的表达明显增加,再灌流3d时达到高峰,14d时接近基础水平。脑缺血再灌流后,大脑皮质CGRP的表达也显著增加,在2h,6h及48h和7d出现三个高峰,再灌流14dCGRP的表达降至基础水平。大脑纹状体CGRP的表达明显增加,再灌流2h即达到高峰,以后反而下降,至再灌流2d时降至基础水平,3d,7d时重新出现高峰,然后14d完全降至正常。脑缺血再灌注后6h开始大脑皮质乙酰胆碱受体的表达升高,至24h达高峰,48h时下降,但仍高于正常(P<0.01),3d,7d时重新出现高峰,然后14d完全降至正常。脑缺血再灌注后2h纹状体乙酰胆碱受体的表达开始升高,48h下降,至3d,7d达到高峰,14d降至正常水平。

【Abstract】 [Objective] To observe the vary regulation of surviving CGRP and AchR, To explore the effects of survivin, CGRP and AchR on brain after damaged .[Metheds] In the present study, immunohistochemistry, HE stain were used in a focal cerebral ischemia- Reperfusion model with middle cerebral artery occlusion in 36 healthy adult Wistar rats to examine the expression profile of surviving CGRP andAchR.[Results] In low magnification of HE stain, neuron in central area necrosis and in semi-sepheral area decreased and degeneration: cell body was triangle, sector, long-strip, with karyopyknosis, deep staining of nucleolus, and neoplasm was vacuoles. Cortex neuron was neoplasm-rarefaction, nuclea-malformation slightly and deep-staining. Degenerated neurons and normal neurons were coexist. There were lots of neurogangalions in both the cetral area and semi-sepheral area. Our results also indicated that the expression of survivin in the cerebral cortex was increased after cerebral ischemiae compared with pseudo-operation. The results showed that the first peak was at 6 hours after reperfusion, decreased at 24h, and then the second peak at 3d, and the second peak was lower than the first one, declined gradually, reached the level of background in 14d. While its expression in striatum increased from 2h to 7d, and the only peak was at 3d, finally reached to background level at 14d. The expression of CGRP in the cerebral cortex was increased after cerebral ischemia compared with pseudo-operation, there was three peaks respectively at 2h and 6h , 48h, 7d, and then decreased gradually to the background level afterwards. While its expression in striatum reached to two peaks at 2h and 3d, 7d. Then decreased gradually to the level of ischemia in initial stage. The expression of AchR in the cerebral cortex was also increased after cerebral ischemia compared withpseudo-operation, there was also two peaks at 6h , 3d and 7d. While in striatum was two peaks at 2h and 3-7d.[Conclusion] The expression of survivin, CGRP and AchR all increased apparently in brain injury, they were related to the re-prosthesis of nevous tissue after damaged.
