凉水、丰林国家自然保护区天然红松林遗传变异的研究
RAPD and ISSR Analysis on Variation of Genetic Diversity of Natural Pinus Koraiensis

【摘要】 本文应用RAPD 和ISSR 技术对凉水和丰林国家自然保护区的天然红松(Pinus koraiensis )居群的共74 个个体遗传多样性变化和遗传分化进行了分析。10 个RAPD 随机引物共检测到61 个位点, 其中多态位点43个, 多态位点比率为69.35%。RAPD 的Shannon指数凉水居群为39.70%,丰林居群为37.35%, Nei 指数统计结果表明, 红松的遗传多样性凉水居群(0.2634)大于丰林居群(0.2607)。13 个ISSR 随机引物共检测到103 个可重复的位点,其中74 个位点具有多态性,多态位点百分比为分别为75.25%和74.00%。ISSR 的Shannon 指数凉水和丰林两个居群分别为43.95%和40.61%,ISSR 的Nei 指数遗传分化为5.63%。RAPD 和ISSR 的两种指数都表明, 红松的遗传多样性凉水居群大于丰林居群,红松种内的遗传变异主要存在于群体内,居群内存在一定的遗传分化。研究表明红松在松科植物中的遗传多样性水平是不低的。由此说明红松物种濒危、分布区逐渐缩小的原因,归根结底是由于人类的破坏作用,再加上火灾和风倒等因素造成的。
【Abstract】 With RAPD and ISSR method,the paper studied genetic diversity and differentiation of two natural population of Pinus koraiensis in Liangshui National Nature Reserve and Fenglin National Nature Reserve. Through the amplification of RAPD with 10 random primers, 61 repeatable loci were detected, and there were 103loci in amplification of ISSR with 13 primers, and ISSR could examine more heredity polymorph than RAPD.(in the RAPD amplification and percentage of polymophic loci was 0.6935%. Nei’s index showed the levels of genetic diversity of Liangshui population(0.2634) was higher than Fenglin’(0.2607); in the ISSR amplification and percentage of polymophic loci was 79.61%.genetic variation existed mainly within populations, there were little genetic differentiation between populations. The level of genetic diversity that the Korean pine is in the loose department plant is not low that studies have shown. Therefore explain why the species of the Korean pine is endangered , the distributing area narrows gradually, it is because of human destruction function after all , in addition, such factors as the fire and wind pour cause.
