薄叶网胰藻Hydroclathrus tenuis和棕色扁海绵Phakellia fusca的化学成分研究

【摘要】 对采自中国南海西沙群岛的薄叶网胰藻Hydroclathrus tenuis和棕色扁海绵Phakellia fusca的化学成分进行研究。从薄叶网胰藻Hydroclathrus tenuis中分离和鉴定出10个单体化合物和1组甾体类混合物,并通过X-ray衍射实验确定化合物1的立体构型,其中化合物1为首次从天然产物中分离得到,化合物2、3、4、5、6、7、8、9、10均为首次从该种海藻中分离得到,化合物10的结构未见文献报道。从棕色扁海绵Phakellia fusca中分离和鉴定出4个单体化合物,其中化合物12、13和15的结构未见有文献报道。 从薄叶网胰藻Hydroclathrus tenuis乙醇浸膏的乙酸乙酯可溶部分分离得到的10个单体化合物,经UV、IR、1HNMR、13CNMR、1H-1HCOSY、HSQC、HMBC、EIMS、HRMS等波谱技术鉴定为:1,4—对卞氧基苯(1),2α,3α,28—三羟基齐墩果—12—烯—24—醛(2),胆甾—4—烯—3β,6β—二醇(3),十六酸乙脂(4),N-(2′-羟基-1′-羟甲基十六烷基)十九脂肪酸酰胺(5),N-(2′-羟基-1′-羟甲基十五烷基)—2,3—二羟基二十四脂肪酸酰胺(6),正十六碳酸(棕榈酸)(7),(25R)-5α-螺甾烷(8),(2S,3R,4R,2′R)-2-(2′-羟基二十二酰胺基)二十六烷—1,3,4—三醇(9),(2S)-2-羟基-N-((2S,3R)-1-β-D-吡喃型葡萄糖基-3-羟基十八烷基-2-十八酰胺10)。从甾体类混合物(11)中鉴定出6个甾醇化合物。 从棕色扁海绵Phakellia fusca浸膏的正丁醇可溶部分分离得到的4个单体化合物,通过经IR、1HNMR、13CNMR、1H-1HCOSY、HSQC、HMBC、ESIMS、GC-MS等波谱技术初步确定为:5,6,7,8.四氢-8-(2-羟基吡啶-3-)-[1,2,3]三唑并[4,5-c]氮杂-4-(3H)-酮(12),N-((2S,3R)-1,3-二羟基三十烷基)—2′S—2′—羟基十二酸酰胺—1″-O-α-D-吡喃型葡萄糖基-(6″→1(?))-α-D-吡喃型葡萄糖苷(13),胆甾—4—烯—3β—醇(14),胍基季铵盐生物碱(15)。
【Abstract】 The chemical constituents of the algae Hydroclathrus tenuis and the sponge Phakellia fusca collected from the South China Sea were studied in the paper. Sixteen compounds have been isolated and identified from the algae Hydroclathrus tenuis. The stereo structure of compound 1 was established by X-ray diffraction analysis. Compounds 1 and 10 were isolated from natural resources for the first time. All the Compounds were isolated from this algae for the first time. Four compounds were isolated and identified from the sponge Phakellia fusca.Ten pure compounds isolated from the algae Hydroclathrus tenuis were identified as 1,4-bis(benzyloxy)benzene(1), (2S,3R,4R)-1,2,3,4,4a,5,6,6a,7,8,8a,9,10,11, 12,12a, 14,14a, 14b-icosahydro-2,3-dihydroxy-8a-(hydroxymethyl)-4,6a,6b, 11,11,14b-hexamethylpicene-4 -carbldehyde(2), (3S,6R)-2,3,6,7,8,9,10,11,12,13,14,15,16,17-tetradecahydro-10,13-dime thyl-17-((R)-6-methylheptan-2-yl)-1H-cyclopenta[a]phenanthrene(3), ethyl palmitate(4), N-(1-hydroxyctadecan-2-yl)nonadecanmide(5), 2,3-dihydroxy-N-(1,3-dihydroxyoctadecan-2-yl)tetracosanamide (6), octanoic acid(7), (25R)-5α-spirostane(8), 2-hydroxy-N-(1,3,4— trihydr- oxytetracosan-2-yl)docosanmide (9), (1-β-D-gucopyranosy-(2S,3R) -2-N-[2R-hy-droxy-octadecyl]-octadecasphinga(10) by UV, IR, 1HNMR, 13CNMR, 1H-1HCOSY, HMQC, HMBC, EIMS, HRMS。Four compouds isolated from the sponge Phakellia fusca were identified as 5,6,7, 8-tetrahydro-8-(2-hydroxypyridin-3-yl)-[l,2,3]triazolo[4,5]azepin-4(3H)-one(12),N-(l,3-dih ydroxytriacontan-2-yl)-2’ -hydroxydodecanmide-1"-O-α-D-glucopyranosyl-(6’’ →1’’’)-α-D- glucopyranoside (13), (3S)-2,3,6,7,8,9,10,11,12,13, 14,15 ,16,17-tetradecahydro-10,13-di methyl-17-((R)-6-methylheptan-2-yl)-1H-cyclopenta[a]phenanthren-3-ol(14), fusca alkloaid(15) by UV, IR, 1HNMR, 13CNMR, 1H-1HCOSY, HMQC, HMBC, EIMS, HRMS。
