藻蓝蛋白与Se(Ⅳ)、Au(Ⅲ)、Ag(Ⅰ)的作用研究

【摘要】 生物大分子修饰的纳米复合粒子成为人们研究的热点。纳米粒子与生物分子相结合具有许多潜在应用,如贵金属纳米粒子与蛋白质结合应用于免疫细胞化学制造等。纳米粒子结合生物大分子能够选择性地与抗原、靶细胞或病毒反应,被广泛应用于免疫标记、生物分子识别、DNA生物传感器和基因治疗等领域,应用非常广泛。 本研究制备了PC-Se(0)、PC-Au(0)和PC-Ag(0)3种复合型纳米缀合物,对其结构及其形成机理进行了研究: 1.藻蓝蛋白(纯度系数A620/A280=3.0,含少量别藻蓝蛋白)与Se(Ⅳ)作用后,藻蓝蛋白溶液的光谱发生了一系列的变化,并在92h后观察到红色纳米硒的生成。这些提示藻蓝蛋白分子是Se(Ⅳ)的主要靶分子之一,硒对螺旋藻的生物效应可能部分是通过硒作用于藻蓝蛋白引起,同时提供了一种制备复合型纳米缀合物的简便易行的新方法。 2.藻蓝蛋白分子可以原位还原Au(Ⅲ)制备纳米Au(0)粒子,形成了PC-Au(0)复合型纳米缀合物,PC-Au(0)复合型纳米缀合物具有很好的分散性和稳定性,PC-Au(0)体系中观察不到普通液相纳米金体系的谱学特性。 3.藻蓝蛋白可以自身原位还原Ag(Ⅰ)制备纳米Ag(0),生成的纳米银基本呈球形,粒径在15~30nm之间,分散于PC表面,没有观察到单独的纳米银粒子存在。生成的PC-Ag(0)复合型纳米缀合物是以PC为核,纳米银为壳部分包覆的复合型纳米缀合物,有很好的分散性和稳定性。 通过研究藻蓝蛋白原位还原制备PC-Se(0)、PC-Au(0)和PC-Ag(0)3种复合型纳米缀合物,有助于对具有抗氧化功能类蛋白质的深入了解,同时,为纳米药物的制备提供了一种新途径。

【Abstract】 Nanoparticles complex modified by biomolecular is playing an increasingly important role.for example, Nanoparticles complex modified by noble metal nanopartical and protein are used in Immunocytochemical production,and so on. Nanoparticles complex modified by nanopartical and biomolecular can react selectively with virus, antigens and target cells.It is used widely in Immuno-labelling, recognition of biomolecules, DNA biosensor and gene therapy.In this paper, the nanoparticles complex of PC-Se(0), PC-Au(0) and PC-Ag(0) were prepared. The structure and formation mechanism of these complexes were also studied.Firstly, after C-phycocyanin(A620/A280=3.0, containing little allophycocyanin) was mixed with Se(Ⅳ), the spectrum of the mixture solution underwent changes and red nano-selenium was observed in the solution after 92h. The information from the spectra of phycocyanin suggested that phycocyanin could be one of the principal target molecules of Se(Ⅳ) and the biological effects of Se(Ⅳ) on spirulina, in part, could be caused accordingly by the interaction of phycocyanin and Se(Ⅳ). And a simple new method of preparing Nanoparticles complex were given.Secondly, PC-Au(0) is prepared from Au (Ⅲ) by in situ reduction of phycocyanin at room temperature without any reducing agent. PC-Au(0) is highly dispersed and stable,but the specific spectra of nano-Au(0) can not be observed on the reduction reaction.And thirdly,PC-Ag(0) is prepared from Ag (Ⅰ) by in situ reduction of phycocyanin at 4℃ without any reducing agent. According to the TEM images, the PC-Ag(0) is a nanostructured bioconjugate with novel core-shell, and the PC-Ag(0) is dispersed and stable and having 15~ 30nm nanoparticles of Ag(0).PC-Se(0), PC-Au(0) and PC-Ag(0) were prepared by in situ reduction of phycocyanin without any reducing agent and the mechanism of reaction were studied. Our results provide some useful inspiration in studing antioxidative protein and give a new method of preparing nano drugs.
