流动注射化学发光分析法在抗菌类药分析中的应用

【摘要】 化学发光分析法(CL)具有灵敏高度、线性范围宽,仪器设备简单等优点,将其与流动注射相结合,更兼有分析速度快,精度高、易实现自动化的特点。由于化学发光法的这些特点,因此这种方法很适合于药物分析,已经被广泛的应用于药物分析领域。本论文包括两部分,第一部分系统评述了该法在药物分析方面的应用,第二部分研究报告,主要依据化学发光原理建立了四种抗菌类药物的化学发光分析法,主要内容如下: (一) 高锰酸钾—阿莫西林化学发光体系测定阿莫西林 在盐酸介质中,高锰酸钾能氧化阿莫西林产生较强的化学发光,据此,采用流动注射技术,建立了一种测定痕量阿莫西林的化学发光分析法。 (二) 高锰酸钾—甲醛—阿莫西林化学发光体系测定阿莫西林 在酸性介质中,高锰酸钾能氧化阿莫西林发生化学发光反应,甲醛的存在可使发光强度增强。据此,采用流动注射技术,建立了一种测定阿莫西林的化学发光分析法。 (三) 流动注射—抑制化学发光法测定阿莫西林 本文基于阿莫西林对CO2+离子催化的鲁米诺-H2O2化学发光体系的抑制作用,首次建立了阿莫西林的流动注射化学发光抑制分析法。 (四) 铈(Ⅳ)—罗丹明6G化学发光体系测定阿莫西林 阿莫西林在NaOH溶液中降解后,其产物可在酸性条件下与Ce(Ⅳ)产生化学发光反应,罗丹明6G对该反应有较强的增敏作用。据此建立了流动注射-化学发光法测定阿莫西林的新方法。 (五) 氨苄西林的化学发光测定法 氨苄西林在NaOH溶液中降解后,其产物可在酸性条件下与高锰酸钾发生化学发光反应,甲醛的存在可使发光强度增强。据此,采用流动注射技术,建立了一种测定氨苄西林的化学发光分析法。 (六) 洛美沙星的化学发光测定法 本实验利用Tb3+能大大增强Ce(Ⅳ)-H2SO3-LMLX的化学发光,采用流动注射技术,建立了一种简单、快速、高灵敏度、高选择性的测定洛美沙星的新方法。 (七) 门冬氨酸洛美沙星的化学发光测定法

【Abstract】 Chemiluminescence (CL) analysis offers high sensitivity , wide linear range and simple instrument. When coupled with flow-injection analysis (FIA), the CL-based FIA methods provide cheap , rapid , simple and reproducible means of detection, and therefore,have been widely used in molecular biologic,biological engineering,medicne, environmental chemistry . The thesis consist of two parts .The first is a brief review commenting the development of chemiluminescence application in pharmaceutical analysis .The other is research reports.The research reports mainly comprise the following eight parts. 1) Chemiluminescent system of amoxicillin-potassium permanganate-hydrochloric acidThe reaction between and potassium permanganate in the sulfuric acid medium gave intensive chemiluminescence.A new analytical method for determination of amoxicillin was proposed. 2 ) An investigation on the chemiluminescence reaction of amoxillin-formaldeh-yde-potassium permangate systemThe chemiluminescence reaction of amoxicillin-formaldehyde-potassium permang-nate system was examined by flow injection technique.A new method for the determination of amoxicillin was developed .3) Flow injection analysis of amoxicillin with inhibited chemiluminescence detectionA new flow injection chemiluminescence method for the determination of amoxillin was described.It was found that amoxicillin inhibited chemiluminescence from luminol-H2O2-Co2+ reaction. On this basis, a f low injection procedure combined with inhibited chemiluminescence detection was developed for the determination of amoxicllin.4) Study and application on chemiluminescent system of degradation product of amoxicillinA new flow-injection chemiluminescence (CL) method for determination of amoxicillin was described .The method was based on CL reaction of the product degraded from amoxicillin in sodium hydroxide medium with Ce(Ⅳ) in acidic condition sensitized by rhodamine6G.5) Chemiluminescence determination of ampicillin by flow injection analysisA novel flow injection the chemiluminescence(CL) method for the determination of ampicillin was described . It was based on CL reaction of the product degraded from ampicillin in sodium hydroxide medium with potassium permanganate . The CL reactionwas sensitized by formaldehyde in acidic solution .6) Determination of lomefloxacin by Tb3+ sensitized chemiluminescenceBased on the Chemiluminescence reaction of Ce(Ⅳ)-H2SO3- LMLX can be sensitized by Tb3+,a simple rapid high sensitive and high selective chemiluminescence method for the determination of LMLX was developed.7) Flow injection analysis of tannic acid with inhibited chemiluminescent detectionLomefloxacin aspartate was found to inhibit chemiluminescence of the luminol-H2O2 reaction catalyzed by Co2+. A flow injection method had been developed for the determination of lomefloxacin aspartate based on the inhibition of the chemiluminescence of the luminol-H2O2-Co2+ system by lomefloxacin aspartate .
