扑草净和表面活性剂OT对蛋白核小球藻的联合毒性研究

【摘要】 本项工作研究了除草剂扑草净与表面活性剂琥珀酸酯磺酸钠(OT)对蛋白核小球藻(Chlorella pyrenoidosa)的单种毒性效应和联合毒性效应。结果表明,无论扑草净和OT是单独使用还是混合使用,低浓度下对蛋白核小球藻均表现出一定的刺激生长的效应,藻细胞密度升高,叶绿素和可溶性蛋白含量的变化与生长曲线极其相似,呈上升趋势,抗体内两种主要的氧化酶SOD和POD活性均上升。随着扑草净和OT的浓度增加,刺激生长效应消失,抑制生长作用明显,叶绿素和可溶性蛋白含量均下降,SOD和POD活性随浓度上升呈先上升后下降趋势,同时丙二醛含量急剧升高,表明细胞内氧自由基大量积累,引起细胞膜脂过氧化,从而导致整体膜的损伤。另外,在单种毒性效应下,扑草净的EC50是O.164mg/l,OT的EC50值是41.380mg/l,而在联合毒性效应中,在两者都在低浓度下,刺激生长现象比两者单独使用时更加明显,二者都在高浓度时则对蛋白小球藻的抑制作用更强:表明不同浓度条件下扑草净和OT表现出相加、拮抗和协同三种不同的效应。
【Abstract】 The single and joint toxicities of prometryne and OT on growth of Chlorella pyrenoidosa were investigated. The chlorophyll content, soluble protein content as well as the density in the green algae cells were increased at the low concentration of prometryne and/or OT, which indicated that the algae growth was stimulated. Meanwhile, the activities of superoxide dismutase (SOD) and peroxidase (POD) were also increased, suggesting that reactive oxygen species was eliminated effectively. With increasing in concentration of prometryne and OT, the chlorophyll content, soluble protein content and the growth of algae were decreased significantly, while the activities of SOD and POD were increased initially, and then conceitedly fell. Additionally, the malonyldialdehyde (MDA) content increased rapidly. The results suggested that active oxygen was excessively and accumulated, and the lipid peroxidant was aggregated, resulting in damage of the algae cell membranes. 96h-EC50 of monotoxicity of prometryne was 0.164mg/l, and OT was 41.380mg/l The growth of algae was faster under both low concentrations than that of each agent alone, while the combinations of both high concentrations were more toxic than expected, which indicated that the both showed concentration addition, antagonistic and synergistic effects under the different concentration.
