数字化细胞微注射中的改性和检测技术

【摘要】 随着生物技术的发展,细胞微操作领域中的细胞微注射技术取得了长足的发展。国内外的研究较多,其中南京理工大学微系统研究室的“微流体数字化技术”取得了突破性进展。“微流体数字化技术”的研究,在基本概念和理论上具有原创性,对微流体系统的研究和微注射技术进步有着重要意义,在细胞微注射技术等方面具有广阔的应用前景。 本论文是上述微流体数字化技术在细胞微注射技术中试验研究的总结。在简要描述微流体驱动控制系统和微注射技术实验条件的基础上,设计了改善微针内外表面特性的疏水化处理工艺,消除了微喷射过程中的渗流、回流现象,改善了微注射过程中的流动特性,提高了微注射效果,得到了数字化微喷射的实验结果;提出了精确测量微针内径的折射率法;设计了纳升级以下的微液滴量的检测方案;最后,论文对微注射技术实验中的原始数据进行了分析。
【Abstract】 With the development of biotechnology, the development of the technology of cell microinjection has been achieved quite greatly in the area of cell manipulation. There are so many researches at home and abroad, while the micro-fluidic digital technology studied in the micro-system lab in Nanjing University of Science and Technology lately has been developed commendably. There are some creative theories and basic concepts in the micro-fluidic digital technology. The micro-fluidic digital technology is uttermost important for the research of the micro-fluid system and the process of the micro-injection technology, and the micro-fluidic digital technology will be applied widely in the area of cell micro-injection.In this thesis, the content based on the micro-fluidic digital technology above the applied research of cell micro-injection is systematically designed. On the base of the brief description of the drive and control system of micro-fluid and the experimental setting of the technology of micro-injection, a novel process of the hydrophobe treating used to mend the characteristic of the inner and outer surface on micro-pipette is designed, which can improve greatly the injected effect and eliminate the disadvantage such as seepage or regurgitation during injection. The author puts forward the way of measuring the inside diameter of the micropipette. The program of measuring micro injection amount is designed. The original data of the micro injection experiment are explained and interpreted at last in the thesis.
