发酵法生产番茄红素及其萃取分离的研究

【摘要】 番茄红素具有淬灭单线态氧、捕获自由基、诱导细胞间信息传递等卓越的生理功能,并具有防癌、抗癌、防治心脑血管疾病等功效。本文主要研究了三孢布拉氏霉菌生产番茄红素的工艺,主要包括:新型阻断剂与促进剂—MPTA的合成及其对发酵的研究,氧载体与表面活性剂对发酵的影响,超临界萃取番茄红素的影响因素等几个方面。 MPTA是一种胺类化合物。在有关类胡萝卜素代谢的研究中,它常作为控制番茄红素向β—胡萝卜素转化的番茄红素环化酶(β-LYC)抑制剂,将代谢控制在番茄红素水平。实验结果表明:采用Williamson反应合成MPTA,收率可以达到70%;在第48h添加1.2mL/LMPTA其阻断效果明显优于在第48h添加烟草废弃物15g/L的对照,番茄红素含量可提高38.3%。 三孢布拉氏霉菌属于高度嗜氧嗜粘度的微生物,发酵生长过程需要大量的氧,尤其是对数生长期溶氧量常常不够,因此我们研究了在发酵过程中添加氧载体及表面活性剂改善溶氧情况。实验结果显示:最优氧载体是正十二烷,在第0天添加1%的正十二烷可使番茄红素产量提高72.84%,在使用氧载体的同时,添加表面活性剂Tween80可以极大的提高番茄红素的产量,与对照相比提高114.83%。

【Abstract】 Lycopene has predominantly functions of quenching singlet oxygen, scavenging of free radicals and reducing the communication between cells, consequently which could act great roles on preventing cancer and atherosclerosis. In this paper, such factors of its technical flow were investigated respectively as follows: synthesis of MPTA as a new bloking agents and accelerant and its influences on fermentation, the influences of Oxygen-vectors, Surface Active Agents on Lycopene’s fermentation, and the effect factors of super-critical carbon dioxide extraction of Lycopene.MPTA is an amine compound, it ws often used to be an inhibitor from Lycopene to β-carotenoids. The experiment said that the enlist could come up to 70% with the help of synthetical MPTA by Williamson reaction.The study indicate that there is 38.3% improvement in the production of lycopene when 1.2mL/L MPTA was added to 2-day old culture of Blakeslea trispora.Blakeslea trispora is highly addicted to aerobic and viscoid medium microorganism, and it need a mass of dissolved oxygen in zymolytic process, especially in logarithm growth phase.
