藏药波棱瓜有效部位化学成分及指纹图谱初步研究

【摘要】 波棱瓜子系葫芦科植物波棱瓜Herpetospermum Caudigerum Wall.的干燥成熟种子,是一种常用藏药,已被中华人民共和国卫生部药品标准藏药(第一册)收载。波棱瓜子味苦,性寒,能清腑热,胆热,具有清热解毒、去火降热、助消化作用,用于治疗肝热、黄疸型传染型肝炎等症。 化学成分预试结果表明,波棱瓜子含有生物碱、氨基酸、蛋白质、多肽、脂肪油、鞣质、酚类、香豆素、木脂素、三萜、甾体、皂苷等成分。袁海龙等从波棱瓜子中分离得波棱素。国外Kaouadji Mourad等人从中分出苯呋喃类化合物Herpetal, Herpetoriol;木脂素类化合物Herpetrione, Herpetetradione, Herpepentol, Herpetol, Herpetetrone等。但该药的药理研究未见报道,其质量控制指标仅有部颁藏药标准波棱瓜子项下的性状鉴别、显微鉴别及生物碱的定性鉴别。 本课题对波棱瓜子抗肝损伤活性部位进行了化学成分研究,从该部位分离出三种单体化合物,经UV、IR、MS、1H-NMR、13C-NMR等鉴定为(Ⅰ)波棱芴酮(herpetfluorenone)、(Ⅱ)波棱酮(herpetone)、(Ⅲ)波棱酚(herpetenol)。三种化合物均系首次从该属植物中得到,且均为新化合物。 以上述三种单体化合物为对照,采用反相高效液相色谱法对波棱瓜子药材进行了指纹图谱研究。选择Alltima C18(250mm×4.6mmID 5um)色谱柱,水-乙腈梯度洗脱,流速1.0ml/min、检测波长230nm,柱温40℃,对10批不同产地批次的波棱瓜子样品进行了测定。研究结果表明,该方法稳定、可靠、重现性好。10批样品得到的色谱指纹图谱有36个共有峰,其共有峰的相对保留时间基本一致,不同产地批次波棱瓜子药材化学组成相似,其相对比例较稳定。
【Abstract】 Herpetospermum seed is the dried ripe seed of Herpetos permum Caudigerum Wall..As a commonly used Tibetan medicinal substance, it is recorded in Drug Standards-Tibetan Medicinal Substances (Vol. 1) ,issued by The Ministry of Health. Herpetospermum seed is bitter in taste and cold in nature. Its actions include clearing heat from fuorgans (e. g.gallbladder), detoxifying, purging the fire, and aiding the digestion. It is indicated for liver heat syndrome ,hepatitis A,etc.The prediction test of its chemical components shows that it contains alkaloids,amino acids, proteins,polypeptides,fatty oil, tannins,phenols, co-umarins, lignans, triterpenes, steroids,saponins,etc. Bolengsu compound was isolated by Yuan Hailong, benzofuran compounds (such as Herpetal and Herpetoriol) and lignans (such as Herpetrione, Herpetetradione, Herpepentol,Herpetol, Herpetetrone) were isolated from this herb by Kaouadji Mourad et al. No pharmacological studies on Herpetospermum seed were ever report. The quality control index of this herb noly include characteristic description, micro-characteristic description and qualitative description of alkaloids in Drug Standards-Tibetan Medicinal Substances (Vol.1)Studying on the chemical constituents from active part for liver diseases of the Tibetan medicinal substance herpeto- spermum seed,three compounds were isolated from it. , By means of UV,IR,MS , 1H-NMR,13C-NMR, they were identified as ( I )herpetfluoren one,
