暴露于二噁英、五氯酚(钠)的男工氧化应激指标、维生素A和生殖激素水平的改变及二噁英对表皮细胞增殖的影响

【摘要】 目的:五氯酚是一种重要的木材防腐剂,其钠盐则广泛应用于血吸虫病防治时杀灭钉螺,其毒性很大。同时在其生产过程中还产生一种毒性更大的杂质二噁英,因此生产五氯酚(钠)作业工人的健康很可能受到这些毒物的危害。本研究从氧化应激、维生素A稳态和生殖激素稳态三个角度探讨生产五氯酚钠的作业环境可能对工人造成的健康影响,为进一步研究提供线索和依据。方法:本次实验以生产五氯酚(钠)的作业男工为暴露组,天津市正常人群作为对照组进行血浆中各项指标的检测,包括氧化应激指标:丙二醛(MDA),谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD);维生索A稳态的指标:维生素A的水平和视黄醇结合蛋白的含量;生殖激素水平改变的指标:睾酮、促卵泡生成素(FSH)和促黄体生成素(LH)。 结果: 1 氧化应激指标改变:与对照组相比,五氯酚(钠)作业工人血浆中丙二醛(MDA)含量明显增高(P<0.01),提示工人体内可能发生了脂质过氧化;同时抗氧化系统中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)均出现下降趋势,但仅有后者的下降才具有统计学意义(p<0.05),。 2 维生素A稳态的改变:与对照组相比,暴露组血浆中维生素A含量有所升高但没有统计学意义(P>0.05),而视黄醇结合蛋白的水平出现明显的上升且具有统计学意义(p<0.01)。 3 生殖激素的改变:与对照组相比,暴露组血浆中睾酮出现明显下降(p<0.01),促卵泡生成素(FSH)则出现明显增加(P<0.05),但促黄体生成素(LH)却未见明显升高(p>0.05)。 结论:长期工作在五氯酚(钠)生产环境中血浆中氧化应激指标出现异常改

【Abstract】 Objective: Pentachlorophenol(PCP) is widely used for the preservation of wood and Pentachlorophenol natrium (Na-PCP) is for the prevention of schistosome, either of which is toxic, besides a kind of dioxins will come up as the inevitable impurities with more toxic during their production. So those workers who had been engaged in the production of the PCP (Na-PCP) are easy to suffer from some short or long health effects as a result of their past exposure to these toxicants. Our present studies will determine if the workers have been influenced in the following three aspects including oxidative stress, Vitamin A homeostasis and reproductive hormones balance to provide further evidences and clues for the prevention and treatment.Methods: In this study the exposed group was defined as those workers who are engaged in the productions of PCP(Na-PCP) and the control group was defined as the unexposed comparison population. A series of assays are conducted in the plasma to obtain three main categories of index: Oxidative stress which was reflected in MDA and SOD and GSH-Px; Vitamin A homeostasis which included Vitamin A and retinol binding protein; Reproductive hormones balance which included testosterone and Luteinzing Hormone and Follicle Stimulating Hormone. Results: First, alteration of oxidative stress index: contrast with that of the control group, the levels of the MDA in the plasma in the exposed group are showed to markedly increase (P < 0.01), which suggested lipid peroxidation had taken place; the levels of SOD and GSH-Px in the exposed group had tended to decrease but the former without statistical significance (P>0.05). Second, alteration of Vitamin A homeostasis: Contrast with that of the control group, the levels of Vitamin A in the plasma in the exposed group are showed to alter without significance (P>0.05), while the retinol-binding protein had exhibited to increase with markedly significance.
