假单胞菌L-21产鼠李糖脂的研究

【摘要】 利用本实验室从油田油污中筛选到的产生物表面活性剂的假单胞菌L-21,对其进行代谢调控的研究,并分离纯化其代谢产物——糖脂类生物表面活性剂。考察了碳源、碳源浓度、氮源、氮源浓度、C/N、金属离子等因素对假单胞菌L-21发酵的影响。并对pH、装液量、接种量、发酵温度和发酵时间等发酵条件进行了研究。假单胞菌L-21以甘油为唯一碳源,30℃,200r/min摇床培养4天,糖脂产量最高达16.50g/L。 假单胞菌L-21的代谢产物经溶剂萃取、柱层析、结晶等分离和纯化得到淡黄色针状晶体,经傅里叶红外(FT-IR)光谱、质谱(MS)和核磁共振(1H NMR、13C NMR)分析鉴定为鼠李糖脂,其主要物质为两种,其分别为Rha-Rha-C10-C10、Rha-C10-C10,并研究了鼠李糖脂的部分性质。

【Abstract】 In this paper, the metabolism of Pseudomonas L-21 was studied and the metabolin was also purified. The effect of medium component and fermentation condition to the Pseudomonas L-21 were studied. Pseudomonas L-21 produced 16.5g/L rhamnolipid at maximum with glycerol as the solo carbon source in shake flask cultures which was incubated at 30℃, 200r/min for 4 days.The rhamnolipid was purified and separated by chloroform-methanol extraction and silica gel chromatography. The rhamnolipid components were characterized as L-rhamnosyl-L-rhamnosyl-β-hydroxydecanoyl-β-hydroxydecanoate and L-rhamnosyl-β-hydroxydecanoyl-β-hydroxydecanoate by FT-IR, nuclear magnetic resonance and mass spectrometry. The part of the properties of the rhamnolipid was studied in this paper.
