聚氨酯功能泡塑分离麻风树种子毒蛋白的研究

【摘要】 改进了麻风树种子毒蛋白粗毒液的制备方法,并通过正交试验确定了粗毒液制备过程中磷酸盐缓冲溶液的最佳条件。实验结果显示,在pH为7.2,浓度为0.02mol/L的100mL磷酸盐缓冲溶液中,麻风树毒蛋白的粗提取率最高,为16.78%。 经过单因素实验,发现当各原料比例为:聚醚100份,甲苯二异氰酸酯150份,二丁基二月桂酸锡0.4份,三乙烯二胺0.6份,硅油1份,水0.1份,聚丙烯酰胺25份,丙酮20份时,采用一步发泡法,使改性聚丙烯酰胺和聚氨酯泡沫塑料高度共混,然后在适当条件下进行胺化反应,合成一种泡孔均匀,弹性良好的吸附泡塑。合成泡塑的某些相关性质为:干视密度为0.11986mg/ml,溶胀度为18%,溶损性为0.0358%。通过其对标准混合蛋白的分离实验,证明了该吸附泡塑对该混合蛋白质具有较好的分离作用。 用功能泡塑对麻风树种子粗毒液进行分离,实验表明,当柱高250mm,用流速为2mL/min的0.02mol/L的磷酸盐缓冲溶液洗脱,麻风树种子中的三组不同蛋白成分能得到较好的分离纯化效果。
【Abstract】 The paper improved the extracting method of crude curcin liquid in Jatropha seeds, and by orthoglnal experiment design , the best condition of the PBS was obtained. The conclusions include following aspects: with PBS of pH=7.2, concentration=0.02mol/mL, volume=100ml, the effiency of extraction of curcin from Jatrapha’ seeds is the optimum, which is 16.78%.By signal method, the best matching of material of foam is found. Under the proportion of the material of foam: Polyether:100, TDI:150, TEDA:0.6, DBTDL:0.4, Si:1, H2O:0.1, PAM:25,acetone:20, with the one step bubble method, the polyamide and polyurethane foam were mixed, then the mixture was treated with amine under the appropriate condition, and the absobate foam with symmetrical bubble and good elasticity was obtained . The related properties of the foam were measured .Its dry density is 0.11986mg/mL, the swelling capacity in water is 18%,the damnify in water is 0.03585. the foam was used to separate the curcin of mixed standard protein,and its result indicate that the foam has good purification.Functional polyurethane foam was used to separate the curcin of Jatropha’ seeds. The experiment indicate, with PBS pH 7.2, column height 250mm,flow velocity 2mL/min, the foam can purify the three kinds of curcin of Jatropha’s seeds well.
