RBP4基因和RARγ基因单核苷酸多态性与小尾寒羊高繁殖力关系的研究

【摘要】 本研究采用PCR-SSCP 方法检测了绵羊视黄醇结合蛋白4 基因(RBP4)和视黄酸受体γ基因(RARγ)的突变位点,并对各基因多态性与小尾寒羊的高繁殖力作了相关性研究。应用PCR-SSCP 技术分析了4 个绵羊品种(小尾寒羊、多赛特羊、湖羊、萨福克羊)中视黄醇结合蛋白4 基因(RBP4)一个位点、5 个绵羊品种(小尾寒羊、萨福克羊、多赛特羊、湖羊、特克塞尔羊)视黄酸受体γ基因的两个位点(外显子Ⅲ、外显子Ⅴ)的遗传变异(基因频率、杂合度、多态信息含量和有效等位基因数)。在各位点中均检测到两个等位基因。RBP4 基因中各群体的杂合度、有效等位基因数和PIC 相差不大,分布比较均匀;RARγ基因外显子Ⅲ位点湖羊和萨福克羊的杂合度、有效等位基因数和PIC 相等并最大,分别为:0.4992、1.9968、0.3746;RARγ基因外显子Ⅴ中特克塞尔羊的杂合度、有效等位基因数和PIC 最大,分别为:0.4838、1.9372 和0.3668。对RBP4 基因和RARγ基因共三个位点在有产羔数记录的小尾寒羊中的多态性的分析表明,每一位点都有两个等位基因,RBP4 基因位点检测到三种基因型,RARγ外显子Ⅲ、和RARγ外显子Ⅴ位点检测到两种基因型。同时分析了这三个位点在有产羔数记录的小尾寒羊中的遗传变异,RBP4 基因为中度多态位点,RARγ基因两个位点均为低度多态位点。方差分析表明RBP4 基因和RARγ基因外显子Ⅴ位点对小尾寒羊的第一胎产羔数、第二胎产羔数有显著影响(P<0.05),RARγ基因外显子Ⅲ位点对小尾寒羊的第一胎产羔数和第二胎产羔数没有显著影响(P>0.05)。由最小二乘均值表明,RBP4 基因第一胎产羔数各基因型的最小二乘均值(LSM1)之间,以及第二胎产羔数各基因型的最小二乘均值(LSM2)之间都是AA 型、AB 型与BB 型差异显著(P<0.05),而AA 和AB 两种基因型之间差异都不显著;RARγ基因外显子Ⅴ位点AA 与AB 基因型均差异显著(P<0.05)。然而RARγ基因外显子Ⅲ位点对LSM1 之间和LSM2 之间的影响都没有达到显著水平(P>0.05)。

【Abstract】 The Polymerase chain reaction-single strand conformational polymorphism (PCR-SSCP) method was used to study the single nucleotide polymorphisms(SNPs) of retinoid binding protein 4(RBP4) gene and retinoic receptor γgene (RARγ) in sheep. Genetic diversity of the RBP4 gene and RARγgene (exonⅢand exonⅤ) was studied in small tail han sheep(xw),dorset sheep(ds),hu sheep(hy),suffolk sheep(sf) and texel sheep(tx) breed especially (except texel sheep for RBP4 gene).Two allele genes were detected in both the RBP4 gene and RARγgene.The heterozygosity, numbers of the effective alleles and polymorphic information content(PIC) were well distributed in the four populations in RBP4 gene, and that were the largest in hy and sf population in RARγgene exonⅢ,which the value was as follow successively: 0.4992,1.9968,0.3746,equally in the two breeds, and that were the largest in tx population in RARγgene exon Ⅴ, which the value was successively: 0.4838,1.9372 and 0.3668。The SNPs of RBP4 gene and RARγgene(exonⅢand exonⅤ) were analyzed in small tail han sheep with lamb record with SSCP method. In this breed, the allele genes were also two for ervery gene site.Three genotypes were detected in RBP4 gene while two in RARγgene exonⅢand exonⅤ.The heterozygosity, numbers of the effective alleles and PIC were also analyzed and the results showed that RBP4 gene belongs to the moderate polymorphism,and the two sites of RARγgene belong to the low polymorphism.No high polymorphism was found in this test. The chi-square test results indicate that the RBP4 gene and RARγgene exonⅤsites affect the first parity and second parity of small tail han sheep remarkably(P<0.05), while RARγgene exonⅢsite has no remarkable effect in the first parity and second parity of small tail han sheep(P>0.05). The Least square analysis of litter size in the first and the second parity showing that: RBP4 genotypes between AA and BB,AB and BB differ significantly (P<0.05),but insigificantly between AA and AB genotype; RARγexonⅤgenotypes AA and AB differ significantly too(P>0.05); while RARγexonⅢseems to have no significant effect on the litter size in the first and the second parity,which may be because there was no difference in fact or the sample was too small, leading to the data didn’t balance.
