厚朴中清除氧自由基主要成份的确定及和厚朴酚延缓衰老作用

Determination of the Main Composition in the Herb of Magnolia Officinalis for Eliminating Oxygen Radicals and the Anti-aging Effect of Honokiol

【摘要】 中药厚:朴的提取物具有很强的抗氧化作用,但其中何种成分起主要作用以及这些成分是否具有延缓衰老的作用目前尚不清楚,为了解决这些问题我们做了相关的研究。 (1) 利用荧光分光光度法测定厚朴酚与和厚朴酚对H2O2和·OH的半数清除率,分别将二者半数清除率与厚朴提取物相应半数清除率相比较可确定厚朴清除氧自由基的主要成分。 (2) 通过测定小鼠心、脑、肝组织匀浆的自氧化和由硫酸亚铁-半胱氨酸激发氧化引起的脂质过氧化产物丙二醛的含量研究和厚朴酚体外抑制脂质过氧化作用。 (3) 通过对高、低剂量和厚朴酚给药组和对照组小鼠的常压耐缺氧实验和负重游泳实验结果的比较研究和厚朴酚的抗应激作用。 (4) 连续42天给小鼠注射0.30g/(kg·d)剂量的D-半乳糖建立亚急性衰老小鼠模型。通过比较衰老组、维生素E阳性对照组、对照组、和厚朴酚高低剂量给药组小鼠血清和心、脑、肝组织中的超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化酶的活性和脂质过氧化产物丙二醛的含量研究和厚朴酚的抗衰老作用。 研究结果表明:厚朴酚与和厚朴酚对羟自由基的半数清除率分别为0.081mg/L和0.091mg/L,对过氧化氢的半数清除率分别为2.357mg/L和2.772 mg/L。计算结果表明:这两种成分在厚朴中的质量分数仅占4.64%,但它们对这两种活性氧的清除作用却在厚朴相应清除作用中占到56.70%和54.91%的份额,由此确定了该两种成分就是厚朴清除这两种活性氧的主要成分。对小鼠的常压耐缺氧实验结果表明,0.002g/(kg·d)剂量和厚朴酚给药组的生存时间可延长16.58%;0.010g/(kg·d)剂量给药组可延长35.66%;都可显著延长小鼠常压耐缺氧的生存时间,并体现出一定的量效关系。小鼠负重游泳实验结果表明,0.002g/(kg·d)剂量和厚朴酚给药组可延长小鼠负重游泳的生存时间31.63%;0.010g/(kg·d)剂量给药组可延长85.98%;同样体现出一定的量效关系。在对小鼠心肌、全脑、肝脏组织进行体外自氧化时,和对照组相比,浓度为2.25×10-4mol/L时对由脂质过氧化产生丙二醛的抑制率在13.57%—70.19%;浓度为1.12×10-3mol/L时的抑制率在44.40%—75.24%。在对同样组织进行半胱氨酸与硫酸亚铁引起的激发态氧化时,浓度为2.25×10-4mol/L时的抑制率在20.53%—78.56%;浓度为1.12×10-3mol/L时的抑制率在34.73%—83.26%;比维生

【Abstract】 The extraction of Magnolia Officinalis possesses the antioxidantive function, but it is not clear which compositions play a main role and whether the compositions possess the antiaging effect. The relational experiments are done to solve the problems.(1) The elimination action of magnolol and honokiol for hydroxyl radical and hydrogen peroxide are determined with fluorescence spectrophotometry, comparing their half elimination ratio with those of Magnolia Officinals, the main efficient composition scavenging oxygen radicals can be determined.(2) The inhibitor effect of honokiol on lipid peroxide is determined by measuring the malondehyde (MDA) of mice tissue (myocardium, brain, liver) in vitro and in Cys-Fe2+ system.(3) Giving mice different dose of honokiol, their antistress effects can be evaluated by measuring the swimming time and the survival time of hypoxia under the ordinary atmosphere.(4) The subacute senile mice model is established by injecting D-galactode 0.30g/(kg·d) for 42 days. All mice are divided into five groups (control group, model group, VE positive control group, high dose group, low dose group). The anti-aging effect of honokiol can be evaluated by measuring the activity of SOD, GSH-Px, CAT and the content of MDA existing in serum, myocardium, brain and liver.The result show the half elimination ratio of magnolol and honkiol are 0.081mg/L and 0.091 mg/L for hydroxyl radical, and 2.357mg/L and 2.772 mg/L for hydrogen peroxide, respectively. The content of magnolol and honokiol in the herb of magnolia officinalis is only 4.64%, however, their elimination action to two kinds of oxygen radicals is up to 56.70% and 54.91% those of magnolia officinalis, so we confirm magnolol and honokiol are the main efficient composition eliminating oxygen radical in the herb of magnolia officinalis. The result of anti-hypoxia experiment show two dose (0.002g/(kg·d), 0.01 g/(kg·d)) of honokiol can obviously prolong the survival time of mice, the prolonging ratio are 16.58% and 35.66% respectively. In the experiment of antifatigue, the positive effect of honokiol is also shown, the expanding ratio of swimming time are 31.63% and 85.98%.
