转基因抗虫棉品种DNA指纹图谱建立和遗传多样性评价

【摘要】 本研究选取国内外陆地棉转基因抗虫棉品种(系)52份,对其纤维品质进行测定研究,建立聚类图;并利用SSR和AFLP分子标记建立52份供试材料的指纹图谱,从表型和分子水平综合分析其遗传多样性。获得主要结果如下: 1.纤维品质分析表明,不同性状在不同品种(系)间存在较大差异。我国转基因抗虫棉不能充分满足纺织工业的要求。纤维长度主要集中在28~30mm,比强度变幅为23.0~31.6cN.tex-1,马克隆值变幅为4.0~5.6,衣分0.36~0.44,纤维比强度变异系数最大。 2.利用13对SSR和AFLP引物,构建了52个转基因抗虫棉的指纹图谱。有32个品种拥有自己的特异谱带,占供试品种的61.5%。在13对引物中,每对引物可鉴别的品种数目不同,没有一对引物可将52个品种(系)鉴别出来。 3.基于纤维品质的遗传多样性分析,欧式距离在0.56~7.92之间,平均欧式距离为3.42,成对品种欧式距离的平均值变化在2.52~5.56之间。类群内平均欧式距离较小。表明品种间有一定的异质性,但异质程度较低。 4.基于SSR和AFLP分子标记的遗传多样性分析,欧式距离在2.00~5.57之间,成对品种欧式距离的平均值变化在3.73~4.75,平均欧式距离为4.21。品种间遗传距离相对较近。 5.利用田间性状和分子标记聚类图均划分为3个类群。在聚类图上,各育种单位的品种交叉分布,品种分类在两种聚类方法中不完全相符。 6.依据表型性状和分子标记分析结果,筛选了中棉所30和邯优778、98N102、DP5432B、DPX680、保铃棉410B六个品种。其中中棉所30相对与其它转基因抗虫棉亲缘关系较远,其它五个品种纤维品质性状较好。
【Abstract】 In this paper, 52 transgenic insect-resistant cotton varieties of Gossypium hirsutum from China and abroad, have been used to investigate fiber quality traits, construct cluster dengrogram. Based on the SSR and AFLP analysis, we analysised the diversity and constructed the fingerprinting of the cultivars. The main results were as follows:1. The analysis of the fiber quality traits showed that there were difference among the experiment materials. The results showed that the transgenic insect-resistant cotton can’t meet the needs of textile industry enough. The fiber length varieties ranged from 28 mm to 30 mm, the fiber strength was within 23.031.6 cN.tex-1, the micronaire was from 4.05.6 and the lint percentage was from 3.64.4. The variance of fiber strength was the biggest among the traits among the coefficients of variance.2. 13 polymophic primer combinations were selected to perform the SSR and AFLP fingerprinting. 32 cultivars have specific bands, accounted for 61.5%. Every primer combination was different in the identification efficiency of cultivars. No primer combination could distinguish all the cultivars.3. The analysis based the fiber quality traits showed that the Euclidean distance was among 0.567.92 and the average Euclidean distance was 3.42. The pairwise Euclidean distance of the varieties ranged from 2.52 to 5.56. It showed that little difference existed among different tested varieties.4. The Euclidean distance was among 2.005.57 and the average Euclidean distance was 4.21 by SSR and AFLP. The pairwise Euclidean distance of the varieties ranged from 3.73 to 4.75 which showed that the genetic basic among the tested varieties is not abundance.5. With the morphological characters, the materials all could be divided into three groups. With the two molecular, the materials all could be divided into three groups too. In every group, there were some cultivars from different breeding provinces. The cultivars in every group were not consistent in two methods.6. Six varieties(lines) were screened form 52 tested materials based on analysis of morphological characters and molecular markers. They were Zhongmiansuo30、
