大豆异黄酮对去卵巢大鼠骨丢失的实验研究

【摘要】 【目的】研究大豆异黄酮(Soybean isoflavones ISO)对去卵巢大鼠骨代谢的影响 【方法】将28只10周龄雌性SD大鼠按体重随机分为四组,每组7只:1、假手术组(Sham-operate SHAM)2、去卵巢组(Ovariectomized OVX)3、去卵巢+大豆异黄酮组(OVX+ISO)予灌胃大豆异黄酮100mg/kg/d 4、去卵巢+苯甲酸雌二醇组(OVX+E)予苯甲酸雌二醇肌肉注射0·1mg/kg一周一次假手术组只切除一小块脂肪组织,其余各组切除双侧卵巢组织。实验期限为12周。12周后股动脉放血处死大鼠,处死前收集血液、24小时尿液来测定血清碱性磷酸酶(ALP)、抗酒石酸酸性磷酸酶(TRACP)及骨碱性磷酸酶(B-ALP)的活性;测定血钙、血磷、白介素—6(IL-6)、尿钙/肌酐、尿磷/肌酐以及尿吡啶酚(PYD)的含量。分离右侧股骨用于骨密度、骨钙、骨磷含量的检测 【结果】去卵巢组大鼠股骨骨密度、骨钙、骨磷含量显著降低(P<0.05),血钙、尿钙/肌酐、尿磷/肌酐、尿吡啶酚均显著增加(P<0.05)。大豆异黄酮和雌激素能明显提高去卵巢大鼠的骨密度、骨钙、骨磷含量(P<0.05),降低血钙、尿钙的含量(P<0.05)。去卵巢组大鼠血ALP、B-ALP、TRACP活性及白介素—6的含量显著增加(P<0.05),异黄酮和雌二醇则降低了大鼠的血ALP、B-ALP、TRACP的活性及IL-6的含量(P<0.05)。 【结论】大豆异黄酮能抑制去卵巢大鼠的骨丢失,促进钙的利用,降低了骨转换,显示了大豆异黄酮的类雌激素样作用。

【Abstract】 [ Objective ] To study the effect of soybean isoflavones on the bone metabolism ovariectomized rats.[Methods] Twenty—eight 10-week-old female Sprague-Dawley(SD) rats were randomly assigned to the following four experimental groups: 1.Sham-operate group (SHAM) 2.Ovariectomized group (OVX) 3. Ovariectomized added soybean isoflavones group (OVX+ISO) Ovariectomized and fed orally soybean isoflavones using a stomach tube (100mg/kg/d) 4. Ovariectomized added estradiol (OVX+E) Ovariectomized and intramuscularly injected with estradiol benzoate (0.1mg/kg once a week ). Rats of the SHAM group only underwent the resection of a bit fat . The rest groups all underwent the resection of bilateral ovaries and fed physiological salinc (NS) on a equal lever with soybean isoflavones . The time of experiment is 12 week. Then all the rats were killed by bloodletting of femoral artery to examine the activities of serum total alkaline phosphatase (ALP), bone alkaline phosphatase (B-ALP) and tartrate resistant acid phosphatase (TRACP). To examine urine calcium , phosphorus compared with creatinine and pyridinoline (Pyd) lever in rats. Separating the right femur of all the rats to examine the mineral density (BMD) and the calcium content of the bone. Results: Rats in the OVX group had significantly lower calcium and phosphorus content and mineral density of the bone (BMD) (P<0.05), and higher serum calcium lever (P<0.05) ,and higher urine calcium and phosphorus compared with creatinine and pyridinoline (Pyd) lever in rats than those in other groups (P<0.05).And serum calcium and urinary excretion of calcium and pyridinoline (Pyd) lever had significantly lower in OVX+ISO,OVX+E groups (P<0.05) , and higher the mineral density(BMD) and the calcium content of the bone in OVX+ISO,OVX+E groups (P<0.05) .
