【摘要】 本文中所研究的中药复方免疫增强剂,经临床应用,能显著提高畜禽成活率、降低发病率,增强机体的免疫功能。以往研究表明,从该复方中提取的复方总多糖能显著增强动物机体的免疫功能,但其作用的机制尚不清楚,本研究欲对该复方总多糖及方中各单味中药多糖的免疫作用及其作用机理作一初步探讨。 分别给小鼠灌服药物(复方总多糖、复方水煎剂、11味单味中药多糖、生理盐水)后制备含药血清,用MTT法测定小鼠淋巴细胞增殖程度,用酶法一氧化氮试剂盒测定腹腔巨噬细胞一氧化氮的生成量,同时与各种药物的体外实验相比较。分别提取各组灌药小鼠脾脏RNA,应用逆转录多聚酶链式反应(RT-PCR)检测IL-2 mRNA的表达水平,凝胶图象光密度分析系统进行相对定量。 结果: 1 复方总多糖、复方水煎剂和11味单味中药的多糖均具有不同程度的促进小鼠淋巴细胞增殖的作用,说明这些药物都具有促进免疫细胞免疫功能的作用。 2 在13个药物血清组中有10组能不同程度地增强淋巴细胞增殖反应,而麦冬多糖药物血清、熟地多糖药物血清、山楂多糖药物血清对小鼠淋巴细胞增殖反应没有增强作用,这可能与有些有效成分未能从消化道吸收,或因其有效成分经消化道或肝脏代谢失活所致。 3 淫羊藿多糖、茯苓多糖、复方多糖、复方水煎剂和黄芪多糖的药物组和药物血清组都能显著增加小鼠腹腔巨噬细胞NO的生成量,说明这些多糖均能显著改善巨噬细胞的功能,增强腹膜的防御功能,对小鼠的非特异性免疫功能有明显的提高作用。 4 复方总多糖、复方水煎剂以及组成复方的11味单味中药多糖均能显著促进小鼠IL-2 mRNA的表达,增强机体的细胞免疫水平,从而说明本实验所采用的中药多糖及水煎剂所产生的免疫增强作用的基础是与其在转录水平增强IL-2 mRNA的表达有关。 5 综合三个实验的结果,发现复方总多糖的免疫增强效果最好,优于复方水煎剂和单一多糖,这可能是因为几种多糖配伍具有协同作用,能更好的发挥细胞免疫增强作用。总体实验结果说明增强淋巴细胞增殖,促进腹腔巨噬细胞NO和脾脏细胞因子的分泌是复方总多糖增强机体免疫功能的部分机制。更多还原

【Abstract】 The compound immunoenhancer of traditional Chinese medicine which studied in this article,can notably increase little animal live rate,lowering the morbidity.strenghen the organism immunity function through clinical application. Research shows that the polysaccharide is the main valid composition of the compound to strengthen immunity function. But the mechanism is not clarity ,this experiment is to notify the immunity function and mechanism of the polysaccharide of Chinese herbs composite and its single Herb of G-Immunoenhancers.Preparation the serum from the total Polysaccharide of Chinese herb recipe and water decoction, each single herb polysaccharide,normal saline enema after oral administration. Determination lymphocyte proliferative and nitric oxide(NO) production of peritoneal macrophages.Reverse transcription-polymerase chain reaction (RT-PCR) and gel documentation analysis system were exploited to detect the IL-2 mRNA expression levels. Results:1 All of the polysaccharides and water decoction can increase lymphocyte proliferate in differ degree,it explains that thesee medicament have effect on immunity function.2 Ten kinds of medicated serum can increase lymphocyte proliferate in different degree, but there are three kinds of polysaccharides of single herbs without such effect, it is possibily caused by some efficiency component can not absorb from the alimentary canal.3 The medicated serum of water decoction and four kinds of polysaccharide, and these medicament itself can notability enhance NO creation,the result means that these polysaccharides can markedness improve the function of M Φ ,strengthen the recovery function of peritoneal of mice.4 All of the polysaccharides and water decoction can increase the expression of IL-2 mRNA,enhance the body’s immunity level.This is one of the mechanism of these medicines.5 Synthetize these results, the total polysaccharide of this composite had the best effect on immune function than all the other medicines. The immunopotentiating effect of total polysaccharide may be associated with its potentiation of IL-2 mRNA expressions at the transcriptive level ,enhance lymphopoiesis and increase creation of NO.更多还原
