VP-16对Tca8113的作用及维拉帕米增效作用的研究

【摘要】 目的:VP-16(etoposide)作为作用于细胞周期S期和G2期的特异性抗肿瘤药物,疗效确切,但国内尚未用于头颈部鳞状细胞癌的化疗。钙拮抗剂维拉帕米(verapamil)能与细胞膜上的P-gp糖蛋白竞争性结合,使化疗药物在细胞内浓度升高,细胞毒效应增强。本研究旨在实验室条件下观察VP-16对舌癌细胞株Tca8113的作用及维拉帕米是能否增强VP-16对肿瘤细胞的抗癌活性,以期临床将VP-16与维拉帕米应用于头颈部肿瘤特别是鳞状细胞癌的化疗。 材料与方法:通过细胞培养技术,在实验室环境中,加入不同浓度的VP-16,采用MTT法研究其对舌癌细胞株Tca8113的细胞毒性作用,并通过末端标记法和流式细胞技术研究其凋亡诱导作用,评价VP-16对颌面部肿瘤的作用。并尝试加入维拉帕米,在不改变其他试验条件下,评价其对VP-16的影响作用。 结果:VP-16可显著抑制舌癌细胞株Tca8113的生长,高浓度组在暴露时间内可大幅度抑制细胞生长,有较高的凋亡率,中低浓度组也有较好效果。维拉帕米本身对Tca8113细胞的生长抑制作用均较小,在0.1,0.5,1.0,2.5,5,10mg/L浓度作用下无明显细胞毒害作用。5mg/L维拉帕米预作用1h可明显提高VP-16的作用效果,其对VP-16有增效作用。 结论:VP-16对舌癌细胞株Tca8113有良好的生长抑制作用和较高的凋亡率,其作用效果与浓度成正相关,增加浓度,可大幅度提高其作用效果。维拉帕米本身的细胞毒性很小,与对照组比较无明显差异,对Tca8113细胞的抑制作用不明显。维拉帕米预处理Tca8113细胞后,明显增加VP-16的作用。

【Abstract】 Objective: VP-16, a special anti-tumor drug which performs the function at the S and G2 periods of the cell cycle, have the exact curative effect. But in domestic VP-16 has no application to the chemotherapy on squamous cell carcinoma of head and neck.verapamil have the ability of the competive combination with the P-gp glycoprotein which can increase the concentration of chemotherapeutics drug in cells and intensify the cytotox effect. We purpose observing the influence of VP-16 on Tca8113,the cells of carcinoma of tongue, and the enhancement effect of verapamil and expect VP— 16 and verapamil be applied better to the clinic chemical therapy on the tumor of head and neck especially squamous cell carcinoma.Methods: in the environment of the laboratory ,the cultured Tca8113 cells were divided into different group. These groups, after taking VP-16 respectively, were detected in the proliferation condition by MTT,TUNEL and FCM, and according these results to evaluate the effect of VP-16 on maxillofacial tumor. We attempt to combine verapamil and VP-16 on Tca8113 cells to evaluate the influence of verapamil on VP-16 without other experiment condition change.Results: VP-16 can markedly inhibit the growth of Tca8113. Compared with control group, there are markedly significant different form the high concentration drug group and the middle concentration drug group.verapamil in itself cannot inhibit the growth of Tca8113 cells,there were no obvious difference between the different drug concentration groups
