白音敖包沙地云杉外生菌根真菌调查及其共生菌土生空团菌纯培养条件的研究

【摘要】 沙地云杉(Picea mongolica)是世界上罕见的树种,也是世界珍稀濒危树种之一,有“生物基因库”和“活化石”之称,集中分布在赤峰市白音敖包沙地。在苗圃条件下,沙地云杉幼苗很难形成菌根,可能是由于苗圃地缺乏相应的菌根菌。为此,本论文主要针对白音敖包地区天然沙地云杉及其主要伴生树种白桦的外生菌根资源状况进行调查。初步调查结果表明:共有7种外生菌根真菌与之共生。即:土生空团菌(Cepococcum geophilum)、美网柄牛肝菌(Boletus,reticulatus)、褐疣柄牛肝菌(Leccinum scabrum)、铜绿红菇(Russula aeruginea)、橙红乳菇(Lactarius akahatus)、疣孢黄枝瑚菌(Ramaria flava)、毒蝇鹅膏菌(Amanita muscaria),隶属5科、7属。通过对采集到的外生菌根真菌子实体、菌根及菌核进行分离、纯化共获得了9个菌株,为实现沙地云杉幼苗的菌根化提供菌种来源。通过调查发现土生空团菌(C.geophilum)是沙地云杉的优势菌根菌。对从沙地云杉小根和菌核上分离得到的两个土生空团菌菌株(分别记为:C.g-ysg和C.g-ysh)进行纯培养条件的研究。研究结果表明:C.g-ysg的最佳培养基为MRD,C.g-ysh的最佳培养基为Pachlewski;C.g-ysg最佳C、N源分别为葡萄糖和硝酸钾,C.g-ysh则为蔗糖和硝酸钾:C.g-ysg在pH值为4~9范围内能正常生长,C.g-ysh在pH值为4~7范围内能正常生长。以上试验结果为土生空团菌的固体扩繁提供了理论依据。

【Abstract】 Picea mongolica is a rare and endangered species in the world, which is conferred to the tide of "biology gene bank" and "living fossil" , and mainly distributes in Baiyinaobao sandy land of Chifeng. Picea mongolica seedlings have difficulty in forming mycorrhizae in nursery, Maybe because there are not corresponding effective ectomycorrhizal fungi. Therefore, a preliminary survey has been performed for ectomycorrhizal resource of Picea mongolica and Betula Platyphylla in Baiyinaobao Region. The survey shows that there are altogether 7 different ectomycorrhizal fungi which are subordinated to 5 families and 7 genera. These ectomycorrhizal fungi are Cenococcum geophilum (C.g), Boletus reticulatus, Leccinum scabrum, Russula aeruginea, Lactarius akahatus, Ramaria flava, Amanita muscaria.By isolating those ectomycorrhizal fungi from fruitbody, mycorrhizae rootlet and scleorotia, we got 9 strains. In the course of investigation, we find that C.g is a predominant ectomycorrhizal fungus in this region. The culture condition of 2 C.g strains which had been isolated from mycorrhizal tissue and scleorotia under Picea mongolica has been tested. These two strains was respectively nominated C.g-ysg and C.g-ysh.The experimental results show that: the best culture media for C.g-ysg is MRD, and the another is Pachlewski; Glucose and KNO3 arc-respectively the best C and N source for C.g-ysg, but the best C and N source for C.g-ysh are respectively sucrose and KNO3; C.g-ysg can grow normally at the range of experimental pH value 49, and C.g-ysh can also grow normally between experimental pH value 47. All these results will provide the theory basis for the reproducing of C.g inoculum.
