肺癌组织中微量元素水平及其形态的研究
Study on Trace Element Levels and Chemical Species in Cancer Tissues and Side-cancer Tissues of Patients with Lung Cancer

【摘要】 本文对19例肺癌患者的肺癌组织和肺癌旁组织、9例肺病良性疾病患者的肺组织中的镉(cadmium,Cd)、铅(lead,Pb)、铬(chromium,Cr)、锰(manganese,Mn)、镍(nickel,Ni)、铜(cuprum,Cu)、锌(zinc,Zn)等七种元素进行了含量分析。结果表明,Cu、Pb含量癌组织高于肺癌旁组织、肺癌旁组织又高于正常组织;Cd、Cr、Mn、Ni、Zn含量肺癌旁组织高于肺癌组织。Pb、Ni可能在肺癌的发生、发展中起重要作用。 肺癌组织、肺癌旁组织经碱性蛋白水解酶消解后,粉末的溶解率分别为70.48%,79.31%。铅和镍的溶出率约3%。绝大多数的铅和镍存在于碱性酶不溶物中。应用X-射线粉末衍射法分析肺粉末酶解不溶物,确定出镍的两种存在形态为氧化镍和单质镍。 酶解液通过Sephadex G-75、Sephacryl S-300HR凝胶分离,结果显示,金属铅在肺癌组织和肺癌旁组织中的化学形态差异显著;在癌组织中形态单一;在癌旁组织中形态多样化。在肺癌旁组织中部分与分子量较小(3,000~70,000)生物分子结合的铅在肺癌组织中转变成了与分子量较大(>70,000Da)生物分子结合的铅。在肺癌旁溶液中部分与小分子量结合的镍化合物,在肺癌溶液中转变成了与大分子量结合的生物分子镍。 本研究证实的肺组织酶解不溶物中大部分镍是以氧化镍和单质镍存在的结果,对揭示肺癌产生中镍元素的作用机理具有重要的意义。
【Abstract】 In this thesis, the levels of Cd, Pb, Cr, Mn, Ni, Cu, Zn in cancer and side-cancer tissues from 19 lung cancer patients and benign tissues from 9 lung patients were performed by using AAS. The results showed that the contents of Cu, Pb in cancer tissues were higher than in side-cancer tissues, and the contents in side-cancer tissues were higher than in benign tissues; Cd, Cr, Mn, Ni, Zn showed lower contents in cancer tissues than in side-cancer tissues. Pb, Ni may play an important role in the oncogenesis and development of lung cancer.Alkaline protease with 1% SDS was used to dissolve the lung powder. 70.48% of cancer powder and 79.31% of side-cancer powder dissolved. Approximate 3% of lead and nickel presented in the supernatant and the others in the residue. X-ray diffraction was performed to analysis the residues. The results confirmed the presence of NiO and Ni.Gel filtration chromatography of the hydrolysate on Sephadex G-75, Sephacryl S-300 HR columns showed a significant difference between cancer and side-cancer tissues. Lead chemical species in cancer tissues were few, whereas lead in side-cancer tissues had a variety of chemical species. Part of lead combined with biomolecules with molecular weight (3,000-70,000) in side-cancer tissues was transformed into biomolecules with higher molecular weight (>70,000Da). Part of nickel combined with molecules in side-cancer tissues was transformed into molecules with higher molecular weight.The results that two species NiO and Ni presented in the residues was very important in revealing the action mechanism of element Ni in the occurrence of lung cancer.
