高压静电场对雏鸡血液和免疫器官免疫变化的影响及机理
Effects and Mechanism of Immunologic Changes on Peripheral Blood and Immune Organs of Chicks Treated with High Voltage Electrostatic Field

【摘要】 本研究应用免疫SPA菌体花环法、间接酶联免疫吸附法、免疫酶组织化学ABC技术及组织化学染色等方法。全面系统地研究了雏鸡经高压静电场(high voltage electrostatic field, HVEF)照射后,其外周血液和免疫器官的体液免疫和细胞免疫的动态变化,旨在揭示高压静电场对雏鸡外周血液及免疫器官的体液免疫和细胞免疫的影响及其机理。研究发现:高压正静电场照射雏鸡后,其外周血液T、B细胞数量,IgG、IgM、IgA含量于照射后1~14天显著高于未照射的对照雏鸡;而高压负静电场照射雏鸡后,其外周血T、B细胞及IgG、IgM、IgA含量于4~14天显著低于相应对照雏鸡。表明高压正静电场在一定时期内能增强外周血液的体液免疫和细胞免疫水平,而高压负静电场对雏鸡外周血液的免疫变化起抑制或负调节作用。高压正静电场照射雏鸡后,其免疫器官胸腺及脾脏TANAE+细胞数、CD4+和CD8+T细胞数及其比值和法氏囊及脾脏IgG、IgM、IgA生成细胞数量于4~21天显著高于对照雏鸡;而高压负静电场照射雏鸡的上述各项指标于7~21天明显低对照雏鸡。提示高压正静电场对免疫器官的免疫机能有促进作用,而高压负静电场则发挥抑制或负调节作用。无论是高压正静电场还是高压负静电场均可引起雏鸡体重明显增加,而高压负静电场使雏鸡增重又显著高于高压正静电场。表明上述免疫指标的变化与体重的增长无相关性。本项研究结果,首次揭示了高压正静电场能增强雏鸡外周血液和免疫器官的细胞免疫及体液免疫水平,而高压负静电场则对其起抑制或负调节作用。同时阐明高压静电场引起机体免疫变化的内在联系及其发生规律,为进一步探索高压静电场引起雏鸡免疫变化的详细机理提供有利数据。实验还发现,高压正静电场与高压负静电场均能引起雏鸡体重增加,高压负静电场较高压正静电场效果更为显著。
【Abstract】 The experiment has applied staphytococcal protein A (SPA) rosette test,indirect enzyme- linked immunosorbent assay(IELISA),avidin biotin-peroxidase complex technique(ABC)and histochemistry staining ways et al. Researching completely into the dynamic changes of the cellular and humoral immunity in peripheral blood and immune organs of chicks after high voltage electrostatic field (HVEF) having irradiated. Intend to discover the effects and mechnisim of immunologic changes on the cellular and humoral immunity in peripheral blood and immune organs of chicks treated with HVEF.The study was as follows: after HVEF irradiated chicks, the numbers of T、B cell and the contents of IgG、IgM、IgA in peripheral blood of chicks by high voltage positive electrostatic field irradiating increased more significantly than control chicks during 1 to 14 day; the numbers of T、B cell and the contents of IgG、IgM、IgA in peripheral blood of chicks after high voltage negative electrostatic field irradiating were more lower than those of control chicks during 4 to 14 day. The results showed that high voltage positive electrostatic field can enhance the level of the cellular and humoral immunity of peripheral blood during some periods , while high voltage negative electrostatic field has inhibition or negative modulation on the immune system of chicks. After HVEF irradiated chicks,the numbers of TANAE+ cells ,CD4+ and CD8+T cell 、its ratios in thymus and spleen and the number of IgG、IgM、IgA antibody-producing cells in bursa Fabricius and spleen of chicks by high voltage positive electrostatic field irradiating increased more than those of no irradiating chicks during 4 to 21 day, while above-mentioned items of chicks by high voltage negative electrostatic field irradiating were fallen more than those of control chicks. From the data can illustrate high voltage positive electrostatic field can greatly strengthen immune function of chicks, on the contrary high voltage negative electrostatic field restrain or negatively regulate it. Whether or not group treated with high voltage positive electrostatic or high voltage negative electrostatic field is irradiated, HVEF brought the increase evidently of body weight of chicks, while the body weight of chicks by high voltage negative electrostatic field irradiating was greater weight than those of chicks by high voltage positive electrostatic field irradiating. It showed that the changes of before-mentioned items have no relation with the increase of body weight of chicks.The above-mentioned results of the study for the first time explains that high voltage positive electrostatic field can advance the level of cellular and humoral immunity in <WP=10>peripheral blood and immune oranges of chicks, while high voltage negative electrostatic field hold back its levels. Meanwhile, it illustrated that HVEF gives rise to the inner relations and regularity on immune changes of chick, in order to further discover the detail mechanism of immune changes of chick treated with HVEF and then provide theoretic basis for relative reaseach. The experiment also discovered that high voltage positive electrostatic field and high voltage negative electrostatic field can bring the increase of body weight of chicks, while high voltage negative electrostatic field has even more evident effect than high voltage positive electrostatic field’.
