胡芦巴化学成分的研究

【摘要】 本研究以传统中药胡芦巴(Trigonella foenum-graecum L.)的干燥成熟种子为原料,采用现代的提取、纯化、分离、鉴定手段,从胡芦巴的乙醇提取物中分离得到11个化合物,经理化常数测定和波谱数据分析鉴定了其中7个化合物的结构。它们分别是小麦黄素—7—0—β—D—葡萄糖苷(Ⅰ)、荭草素(Ⅱ)、牡荆素(Ⅲ)、槲皮素(Ⅳ)、胡萝卜苷(V)、十八碳酸(Ⅵ)、β-谷甾醇(Ⅶ)。其中化合物Ⅱ、Ⅶ为首次从国产胡芦巴中分得。对胡芦巴化学成分提取工艺进行比较研究,最佳工艺为乙醇提取法:浓度为85%,提取2次,乙醇用量为10、8倍,提取时间每次各为1小时。采用不同型号的大孔吸附树脂对胡芦巴提取物通过正交试验进行了纯化工艺的优选研究。结果表明:树脂法的富集纯化效果优于乙醇提取法;以D101的富集纯化效果最好。最佳纯化工艺为:D101树脂柱,树脂:药材=1:1,药液浓度=30%,药液温度:70℃。采用HPLC法对不同产地胡芦巴中薯蓣皂苷元的含量进行比较研究,结果表明:河北产胡芦巴中薯蓣皂苷元的含量明显优于其它产地。采用GC法对不同产地胡芦巴中亚油酸的含量进行比较研究,结果表明:河北产胡芦巴中亚油酸的含量明显优于其它产地。通过药理活性实验研究证明胡芦巴乙醇提取物对葡萄糖性小鼠高血糖有明显的降血糖作用,而且胡芦巴乙醇提取物对S180模型小鼠有较好的抗肿瘤活性。

【Abstract】 In this research 11 compounds were isolated from the seeds of Trigonella foenwn-graecum L. by modern technologies of extraction, isolation, purification,identification,such as macroporous absorption resin(MAR),silica gel-column chromatography,sephadex LH-20 and Polyamide column chromatography.The structures of 7 compounds have been established and identified on the basis of physico-chemical and spectrographic evidence.They were tricin-7-O- -D-glucopyranoside , orientin , vitexin , quercetin , daucosterine , stearic acid, -sitosterol.The different optimum extraction conditions of Trigonella foenum-graecum L. were determined by the orthogonal test and the content of total saponins with diosgenin as index was detected by the UV spectrophotometry so as to compare the traditional extraction method by ethanol with the modern method by MAR.The result indicates that the extraction enriched by D101 MAR can enhance the content of total saponins obviously.The research detected the content of diosgenin in the extraction and linoleic acid in the lipid of samples of different habitats by HPLC and GC respectively.And the results show that the content of these two indexes are highest in the sample of Hebei province.Through the pharmacological experiments the results prove that the extractions of Fenugreek seeds exhibit a significant hypoglycemic and anticancer activity.
