丹参组织培养技术研究及组培苗生产试验

A Study on Technique of Tissue Culture and Field Experiment Using Test-tube Nursery Stocks in Salvia Miltiorrhiza Bunge

【摘要】 通过从1999到2002四年对中江大叶丹参进行组培技术研究及组培苗生产比较试验,建立了中江大叶丹参的快繁技术体系,并评估了组培苗的生产性能。结果表明: 1.中江大叶丹参的0.2—0.3cm芽在1/2MS+6-BA0.4mg/L上成活率最高,生长正常。 2.中江大叶丹参温室叶片和试管苗叶片的诱导培养基以1/2MS+6-BA3mg/L最好。温室植株叶片比试管苗叶片出芽晚,但最终两种不同来源的叶片的出芽率不存在差异 3.8~11月和2~4月两个季节进行继代增殖,1/2MS+6—BA0.4mg/L是中江大叶丹参的继代增殖的最佳培养基,2~4月接种的苗子,增殖倍数最高达6.03倍,而6~8增殖倍数只有3.57倍。 4.继代增殖的芽苗在2/3MS+6-BA0.1mg/L上进行壮苗,以备生根。 5.瓶内生根试验中,壮苗在2/3MS+IBA0.2+NAA0.2上10天生根率达98%,且长势好,未经壮苗的苗在20天时生根率只有68%。瓶外生根试验中,壮苗经过50mg/IBA和NAA的混合液浸泡基部后成活率达75%,而未经壮苗的苗在75mg/L的IBA和50~75mg/L NAA混合液浸泡基部后成活率只有52~55%。 6.生产试验证明,组培苗比对照苗的生药产量高,且生长势较好。组培苗药用成份中丹参酮Ⅱ_A的含量高达2.54%,平均比对照高20.39%。

【Abstract】 Rapid propagation technique system of Salvia miltiorrhiza Bunge was established by the study of its tissue culture from 1999 to 2002, and field experiment proved that the quality of the plantlets was superior and quantity were improved. The results were as follows:1. The highest survival of 0.2-0.3cm shoots from Salvia miltiorrhiza Bunge was observed onl / 2MS medium with 6-BA0.4mg/L.2. The best medium for regeneration of leaves from test tube plants and greenhouse plants was 1 / 2MS medium with 6-BA 3mg/L .the leaves from greenhouse plants regenerated later compared with those from test tube plants, but the inductive rates of adventitious shoots from both kind of leaves were not significantly different.3. The best sub-culture medium for Salvia miltiorrhiza Bunge was 1 / 2MS medium with 6-BA0.4mg/L.Multiplication rate differed from season to season: the explants cultured from February to April resulted in a multiplication rate of 6.03, whereas those cultured resulted in a multiplication rate of 3.57 from September to November.4. Using 2 / 3MS medium containing O.lmg/L 6-BA ,stronger plantlets could be produced.5. In in vitro rooting trial, the rooting rate of plantlets was 98% on 2 / 3MS4- IBA0.2 + NAA0.2 after 10 days compared with 68% after 20 days. In ex vitro rooting trial, survival rate of plantlets was 75% by dipping their stem base in solution containing 50mg/IBA and NAA compared with 52~55% by dipping their stem base in solution containing 75mg/IBA and NAA.6. The field trial proved that the seedlings from tissue culture were superior to the ordinary seedlings by cutting. The content of Tanshinone IIA in crops from tissue culture seedlings reached 2.54%, 20.39% higher than that of contrast.
