小麦抗纹枯病基因的遗传和分子标记分析

【摘要】 本研究以RIL群体为材料,进行了小麦抗纹枯病基因的遗传分析。构建了RIL-8群体的分子标记遗传图谱,并对纹枯病进行了QTL分子标记定位。对RIL-SES群体的抗纹枯病基因进行了分子标记分析。(1)利用15个小麦重组自交系群体(RIL)对抗纹枯病进行了遗传分析,结果表明,所分析的群体全部是由主基因和微效基因共同控制;在2对主基因存在时,都有主基因间的互作效应,其值一般小于加性效应;主基因对变异的贡献大于微效基因,主基因的作用更为重要。(2)利用RIL-8群体进行了分子标记分析,共拟合42个SSR标记多态性位点。综合李斯深等绘制的该群体图谱,共拟合94个SSR标记位点,31个ISSR位点,3个HMW-GS位点,涉及20条染色体(不含3A)。(3)对RIL-8群体纹枯病抗性进行了QTL检测,检测到一个加性QTL,位于1A染色体上,贡献率为21.57%; 检测到4对QTL间互作位点,涉及7条染色体,互作贡献率分别为11.63%、6.54%、14.04%、20.01%,互作总贡献率为52.23%。(4)通过对RIL-SES群体118个系(从196个系中随机选择)检测,发现一个SSR标记Xgwm526,位于2B染色体上,与纹枯病抗病基因距离为27.9cM;一个ISSR标记IS840,与纹枯病抗病基因距离为16.9cM。
【Abstract】 In this study, RIL populations are used to analyse the inheritance of sharp eyespot resisitant gene in wheat. The linkage map of SSR and ISSR molecular markers is completed using RIL-8 population. The linkage-ship between molecular markers and sharp eyespot resistant gene was analysed using RIL-SES population.The main results are as follows.15 RIL populations are used to analyse inheritance of sharp eyespot resistant gene. The resistant gene is controlled by major genes and mutipple genes. When two major genes exist, they always exsist epistatic effects which values are generally smaller than additive effects. Contribution of major gene is greater than mutipple gene, so major gene is more important. Based on the RIL-8 population, a genetic map including 42 SSR locis was constructed. Adding Li’job, a genetic map including 94 SSR locis, 31 ISSR locis and 3 HMW-GS locis distributing 20 chromosomes (not including 3 A) was constructed.Based on the genetic map with 128 distributed molecular marker locis, we indentified 1 addative and 4 pairs of epistatic quantitative trait loci(QTLs) contributing to sharp eyespot resistance. The addative QTL locating 1A chromosome could explain 21.57% of the total phenotypic variation. 4 pairs of epistatic QTLs which interaction effect values were 11.63%、6.54%、14.04%、20.01%, locate on 7 chromosomes , and their total interaction effect value was 52.23%.RIL-SES population with 118 individuals was used to indentify molecular markers linked to sharp eyespot resistant gene. As a result, 2 molecular markers, including 1 SSR and 1 ISSR marker, were detected linked to sharp eyespot resistant gene. This dominant resistant gene was mapped on chromosome 2B. The linkage distance between the markers was 27.9cM and 16.9cM.
