高效反硝化菌的实验研究及其脱氮动力学分析

【摘要】 在当前经济发展过程中,水资源紧缺、水环境日趋恶化已经成为制约我国经济发展的重要因素,水体污染的治理及防治已迫在眉睫。生物反硝化法是解决水体中硝氮污染问题的主要方法之一。本实验采用实验室筛选出的高效反硝化菌YZ-2,研究了不同条件对YZ-2进行生物反硝化的影响。实验分别采用蔗糖、葡萄糖、混合碳源及柠檬酸钠为碳源进行了间歇实验。首先,通过改变pH值及温度,进行一系列间歇性实验,确定了反硝化进行的基本条件;其次,对单一碳源及混合碳源(蔗糖、葡萄糖)在室温及低温下的反硝化效果进行了对比。结果表明,菌株YZ-2具有很强的反硝化能力,经YZ-2处理过的污水可以达到国家标准。为了对反硝化菌YZ-2的性能有进一步的了解,通过Monod方程进行了动力学分析,得出了如下模型:COD降解Monod模型、硝酸盐氮反硝化Monod模型、温度对动力学的影响以及pH值对动力学参数的影响的模型;本实验还采用海藻酸钠和聚乙烯醇对菌体进行包埋固定,制成菌球,通过连续性实验,进行了固定化菌球降解硝氮废水的研究。通过考察实验条件对菌株反硝化效果的影响,确定了最佳的工艺参数。
【Abstract】 In the course of present economic development, water scarcity and water deterioration have became important limiting factors of national economic developments in our country. The remediation and precaution of the water pollution have been extremely urgent.Biologic denitrification is one of the most popular methods to remove nitrite and nitrate from water. In the experiment, we used the dentrifier named YZ-2 which has been isolated in the laboratory. We researched the influence of different conditions on the dentrifier. In the batch experiment we used the sucrose、glucose, mixed carbon resource and sodium citrate as the carbon resource. First, through changing the pH value and temperature, we carried on a series of batch experiments and the primary condition have been determined; Secondly, we compared the denitrification result of single carbon resource and mixed carbon resource (sucrose, glucose) under room temperature and low temperature respectively. The result indicates, denitrifier YZ-2 has very strong denitrifying ability. The level of water can reach the national standard after treatment with of YZ-2.In order to have further understanding on the properties of YZ-2, Monod equation is employed to achieve kinetics analysis in this study. And four mathematical model have been received such as COD degradation Monod model, nitrate denitrification Monod model, the model of influence of temperature on dynamics and the model of effect of pH value on dynamics parameters. Moreover, sodium alginate and polyvinyl alcohol are used to embend and immobilize the denitrifier, meanwhile, the research on degrading the nitrogen waste water by the immobilized cell has been carried out through continuous experiment. Finally, the optimal technological parameters are defined by way of observing and studying the influence of experimental condition on denitrification.
