丝裂原活化蛋白激酶在非小细胞肺癌中的表达及其临床意义

【摘要】 目的:探讨丝裂原活化蛋白激酶在非小细胞肺癌中的表达及其临床意义。 方法:手术切除52例非小细胞肺癌及癌旁肺组织标本,应用固定化蛋白质印迹法检测肺癌及癌旁正常肺组织中MAPKs的表达情况;应用免疫组织化学法分析其在组织和细胞内的定位。 结果:肺癌组织中ERK1、ERK2、P-ERK1/2、JNK、P-JNK及P-P38蛋白的平均表达水平明显高于癌旁正常组织,分别为癌旁组织的1.5、1.8、1.7、2.3和2.1倍(P值均小于0.01)。其中P-P38蛋白表达水平在52例肺癌标本中全部高于癌旁正常组织,差异有显著性统计学意义,而P38蛋白的表达水平与癌旁组织无明显差别(P>0.05)。进一步研究表明P-P38与非小细胞肺癌的病理分型、肿瘤大小、分化程度、淋巴结转移、TNM分期及吸烟与否均无关。免疫组织化学表明,ERK1、ERK2、P-ERK1/2、JNK、P-JNK及P38仅表达于细胞浆内;而P-P38在胞浆和胞核内均有表达。 结论:MAPK通路与非小细胞肺癌的发生、发展密切相关。P38的过度激活在非小细胞肺癌的发生、发展过程中可能起着重要的作用。
【Abstract】 Objictive: To investigate the expression and activation of mitogen-activated protein kinases in non-small cell lung cancer and its clinical significance.Methods: Western blot was used to detect the expression of MAPKs (ERK1, ERK2, P-ERK, INK, P-JNK , P38 and P-P38) protein in the surgically resected in 52 patients with non-small cell lung cancer. Immunohistochemistry was used for localization of MAPKs protein.Results: The levels of ERK1, ERK2, P-ERK1/2, JNK, P-JNK and P-P38 protein in non-small cell lung cancer tissues were increased,which were 1.5, 1.8, 1.7, 2.3, 2.1 and 2.1 times compared with those in adjacent normal tisses respectively,and they all have remarkable difference(P<0.01). P-P38 was increased in all 52 cancer tissues, but there were no difference of the level of P38(P>0.05) between cancer tissues and corresponding normal tissues. Further study showed that the expression of P-P38 was not correlativity with tumor size,TNM staging,lymph node involvement, differentiation,pathologic type or smoking. Immunohistochemistry showed that ERK1 , ERK2, P-ERK, JNK, P-JNK and P38 protein were mainly located in cytoplasm and P-P38 were located in both cytoplasm and nucleus.Conclusions: MAPK signal transduction is correlative with human non-small cell lung cancer closely. The overactivity of P38 may play an important role in the development of human non-small cell lung cancer.
