流动注射分析系统的开发及氨基酸的快速检测研究

【摘要】 虚拟仪器是90年代以来随着计算机技术的进步而逐渐发展起来的一类新仪器产品,是指具有虚拟界面,基于计算机的数据采集测试分析仪器。本文从流动注射分析实际需要出发,设计开发了一套化学虚拟仪器系统。 在综合研究了国内外虚拟仪器发展、设计理论、开发方法的基础上,比较了各种仪器的测试原理及其适用条件,确定了系统的设计方案。 本套虚拟仪器采用PCI总线卡式结构,数据采集部件采用具有AD、DI、DO功能的多功能数据采集卡,溶液处理采用蠕动泵、旋转阀,检测器采用分光光度计。软件开发采用Visual Basic 6.0,根据流动注射分析实际需要定义软件功能。 整套系统界面友好、操作简便、功能较完善、运行稳定、响应灵敏、集成化程度高、维护方便,而且为其它分析仪器的微机化提供了硬件和软件平台。运用虚拟仪器设计思想,整个开发工作周期短,难度小。 最后利用此分析系统进行了氨基酸总量快速检测方法研究: 以茚三酮为显色剂,采用流动注射停流光度分析法,研究了酸度、载流流速、采样体积、加热时间等条件对氨基酸测定的影响,得到最佳测定条件:pH=5.4,λ=570nm,载流与试剂流速为2.0ml/min,显色剂浓度为0.2%,加热时间6min。氨基酸浓度在0~2.0μg/ml范围内与其吸光度呈线性关系。 该方法分析精度高,试剂耗量小,操作简单,大大缩短了分析时间,可根据生产实际需要设计成专门的氨基酸总量测定仪,并用于生产过程质量控制。

【Abstract】 Virtual Instrument is one kind of new instrument that has developed step by step along with the progress of computer technology since 1990, which is one kind of instrument that collects data and analyzes data by computer, it has visual panel. This thesis discussed how to design one set of chemical Virtual Instrument to fulfill the need of Flow injection Analyze.The Project was designed by comparing the theory and application area of all kind of instruments based on researching the progress of develop, the theory of design and the method of execution of visual instrument in and out of our country.This visual instrument works on common circuit of Peripheral Component Interconnect, it is made up of data acquisition card, peristalsis pump, injecting sample valve and photometer. The data acquisition card has the function of AD, DI and DO. The peristalsis pump and injecting sample valve were used to deal with liquid. The photometer was used as detector. The software was programmed with Visual Basic 6.0, it function was designed according to the need of FIA.The interface of the system is friendly. It is simple and convenient to operate the system. The function of the system is perfect .The system can work stably and sensibly. Because it is made up of integrated component, it is convenient to maintain it. Other apparatus also can use the hardware and software of this system.At last, this thesis discussed the research of method of determine the total content of amino acid quickly.Employed with ninhydrin and FIA, the acidity, the velocity of carry liquid, the volume of tube of gathering sample, the time of heat the liquid and other parameter that affect the result of determining of amino acid were discussed and the best condition was gained: pH=5.4; =570nm; the velocity of carry liquid and reagent is 2.0ml/min; the content of ninhydrin is 0.2%;the time to heat the liquid is 6min.It has a good linearity of the content of amino acid and its absorbency when the content of amino acid is between 0~2.0 g/ml.The precision of this determine method is good, and the wastage of reagent is a little. It is easy to apply the method and it can shorten the time of determining by a long way. According this method, we can design the special instrument to determine the total content of amino acid and apply it to quality control in factory.
