重组人生长激素对腹腔化疗大鼠骨髓影响的实验研究
The Effect of Recombinant Human Growth Hormone on Bone Marrow Hematopoietic Function in Intraperitoneal Chemotherapied Rats

【摘要】 目的 探讨重组人生长激素(rhGH)对化疗大鼠骨髓的影响。方法 应用血液自动分析仪、骨髓涂片、病理切片及免疫组化法(PCNA)检测实验大鼠外周血和骨髓中各系细胞的变化。结果 给药后第7天外周血中rhGH组网织红细胞(Ret)绝对数较各组明显增加(P<0.05)。化疗组和化疗+rhGH组白细胞(WBC)数、血红蛋白(Hb)含量和Ret绝对数明显降低(P<0.05),但后者WBC和Ret绝对数减少程度低于化疗组(P<0.05)。骨髓形态学检查,化疗组和化疗+rhGH组骨髓增生极度低下或明显减低,骨髓有核细胞总数显著减少(P<0.05),非造血细胞比例明显增加(P<0.05),但化疗+rhGH组增加程度低于化疗组(P<0.05),并可见骨髓小粒、红细胞嗜多染及少量巨核细胞。第14天化疗组和化疗+rhGH组外周血中红细胞数(RBC)和Hb含量明显下降(P<0.05),但化疗+rhGH组较化疗组下降程度低(P<0.05),并且Ret绝对数较各组明显增加(P<0.05)。WBC数各组间比较无统计学意义(P>0.05);化疗组和化疗+rhGH组骨髓增生极度活跃,后者骨髓有核细胞总数较化疗组明显增加(P<0.05),两组各系比例恢复正常。第21天,化疗组RBC、Hb计数低于对照组和rhGH组(P<0.05),化疗+rhGH组RBC计数较rhGH组低(P<0.05),而Ret绝对数高于各组(P<0.05)。第7、14、21天化疗组PCNA阳性细胞数均明显低于rhGH组和化疗+rhGH组(P<0.05)。结论 rhGH对化疗大鼠的骨髓抑制有一定的保护作用。
【Abstract】 Objective To investigate the effect of recombinant human growth hormone (rhGH) on bone marrow hematopoietic function in chemotherapied rats. Methods Analysis of peripheral blood by automatic blood cell counter, smearing for bone marrow cells, pathological section for bone marrow and the expression of proliferating cell nuclear antigen (PCNA) in bone marrow by immunohistochemical method Results Day 7 after administration, reticulocyte count had obviously increased in rhGH group (P<0.05), while the white blood cell (WBC) count, hemoglobin and reticulocyte count was decreased distinctly (P<0.05) in chemotherapy group and chemotherapy+ rhGH group, however, the decreased degree in chemotherapy+ rhGH group was less than that in chemotherapy group (PO.05). in myeloid morphology, myeloid cell was hypoplastic excessively hi chemotherapy group, and it was hypoplastic obviously in chemotherapy+ rhGH group. Myeloid karyote count was dropped significantly and the proportion of non-hcmatopoietic cell was raised distinctly both in chemotherapy and chemotherapy+ rhGH group (PO.05). But there were no myeloid granules and few megakaryocyte in chemotherapy group, while some myeloid granules, megakaryocyte and polychromatic erythrocyte could find in chemotherapy+ rhGH group. Day 14, the counts of red blood cell (RBC) and hemoglobin were dropped distinctly in chemotherapy group and chemotherapy+ rhGH group (P<0.05), but the decreased degree in chemotherapy group was more thanthat in chemotherapy rhGH group (P<0.05). Reticulocyte count of chemotherapy+ rhGH group was higher than others (P.005). There was no statistical significance in WBC count (P >0.05). In myeloid morphology, myeloid cell was hyperplastic actively excessively in chemotherapy and chemotherapy rhGH group, and myeloid karyote count was increased significantly in chemotherapy+ rhGH group than that in chemotherapy group (P<0.05), and morphology of myeloid series was recovered. Day 21, RBC and hemoglobin count hi chemotherapy group was less than control group and rhGH group (PO.05), and hi chemotherapy rhGH group the counts of RBC was lower than rhGH group (P<0.05), while reticulocyte count was higher than others still (PO.05). Day?, 14 and 21, PCNA positive cells count hi chemotherapy group was lower than rhGH group and chemotherapy rhGH group (PO.05). Conclusion rhGH has a protective effect on myeloid hematopoietic function hi intraperitoneal chemotherapied rats.
