樟树优良单株选择与组培研究

On Superior Tree Selection and Tissue Culture of Cinnamomum Camphora

【摘要】 本文通过对南平、三明、龙岩等地樟树种质资源的调查与分析,将该地区的樟树按照主要化学成分划分为四种化学类型:芳樟型、脑樟型、桉樟型以及不明物型。在对前3种化学型樟树的叶片进行切片光学显微镜和扫描电镜观察时发现,其叶片显微结构存在一定差异。参照国际市场芳樟醇产品的标准,经过统计分析确定了芳樟优树的选择标准,从已测定的大量芳樟单株中选出了5株各项指标都达到优树选择标准的芳樟优树。 利用选出的芳樟优树为母树,从外植体选择到成苗的整个过程,研究了影响芳樟组培的各个环节。选出了最适宜的外植体为第3节以下嫩梢茎段,最佳的启动培养基为MS+BA1.0+IBA0.1(或NAA0.1)。增殖培养实验表明:(1)从基部辐射状萌发多个嫩枝和从主茎的腋枝萌生两种增殖方式同时发生有利于提高增殖率;(2)通过试验,筛选出最佳增殖培养基为MS+BA3.0+IBA0.2,在增殖培养基中培养45天,平均增殖系数达到6.1;(3)将培养基的琼脂浓度提高至6.0g/1,pH值调至6.0等手段,继代增殖过程中严重的玻璃化现象得到有效缓解;(4)加入抗氧化剂,选择合适的外植体材料以及缩短继代周期对于减轻褐化有一定效果。采用瓶内生根和瓶外扦插生根两种试管苗生根方法培养时发现,后者生根率和成苗率都很高;苗基部蘸1000ppmIBA,扦插于上层蛭石、下层珍珠岩(体积比1:1)的基质中,成活率81.9%,生根率91.6%,而且这种方法可简化培养程序,缩短成苗期。

【Abstract】 The genetic resources of Cinnamomum camphora distributed over the area of western Fujian (Nanping, Sanming and Longyan), China, were investigated and analysed. In terms of its chemical composition, four types of Cinnamomum camphora were identified, Linalool, Camphor, Cineol and The unknown. The microscopic structures of the leaves collected from the former three types of trees were found to be different under the optical and electron microscopes. With reference to the standard of linalool products in the international market, the selective criterion for superior trees was determined through statistical analyses and consequently five superior trees were selected for the experiment.The factors influencing tissue culture of the selected superior trees were studied, indicating that the optimum explants are the top tender stems under the third nodes, and the optimum initial medium is MS+BA1.0+IBA0.1(or NAA0.1). The propagation culture experiments revealed: (1) The simultaneous occurrence of the two propagation ways, shooting radially from the base of the stems and the axillary shoots of the main stems, is beneficial for elevating the propagation rate; (2) The optimum medium selected from the experiments is MS+BA3.0+IBA0.2, with the average propagation coefficient reaching 6.1 after being cultured in the medium for 45 days; (3) When agar concentration and pH values of the medium were adjusted to 6.0 g/l and 6.0, respectively, the severe vitrification in subculture propagation process was effectively alleviated; (4) By adding antioxidant VC to the medium, using suitable explant materials, and shortening subculture cycle, brownification can be mitigated to some extent. Compared with rooting intube, cuttage rooting in transplantation medium exhibited higher rooting rateand seedling survival rate. When treated with 1000 ppm IB A and planted in amedium with perlite on the top and vermiculite on the bottom (volume ratio:1:1), the cuttings had a survival rate of 81.9% and rooting rate of 91.6%,suggesting it is a quick and practical way for tissue culture.
