中国林蛙皮肤活性肽分离提取的研究

【摘要】 中国林蛙(Rana Chensinensis)是我国重要的经济蛙类,也是黑龙江省优势药用价值动物资源之一。目前,国内针对中国林蛙的研究多见于雌性中国林蛙输卵管——蛤士蟆油的成分和功能等方面的报道,而对中国林蛙皮肤活性物质的研究少有报导。蛙类皮肤中含有丰富的具有特殊分子结构、功能复杂多样的生物活性物质,具有重要的应用价值及较大的开发利用潜力。着眼于疾病治疗因子的筛选和活性分子的发现,如能从中国林蛙皮肤中分离提取出具有重要价值的生物活性物质,必能丰富“北药”资源,为更有效的开发利用中国林蛙资源打下坚实地基础。 本试验选择黑龙江省帽儿山产中国林蛙的皮肤为研究材料,提取皮肤活性肽,并通过对提取体系、抽提温度、抽提时间等因素进行了筛选优化,确定了活性肽的最佳提取条件和技术路线。本试验摸索出用SDS-PAGE电泳分析小分子多肽的电泳条件,并建立了用聚丙烯酰胺凝胶电泳法分离提纯活性肽的实验方法。 本试验通过抑菌试验检测林蛙皮肤活性肽的生物活性,菌种为:大肠埃希氏菌、金黄色葡萄球菌、绿脓杆菌、放线菌。试验结果表明:林蛙皮肤中小分子活性肽对革兰氏阳性细菌、革兰氏阴性细菌具有一定的抗菌活性。说明中国林蛙皮肤活性肽具有良好的开发利用前景。

【Abstract】 Chinese Forest Frog (Rana chensinensis) is a kind of important amphibian in China and also one of the most important medical animals of Hei Longjiang Province. At present, there are plenty of reports on the contents and functions of the Oviductus Ranae about the study of Chinese Forest Frog, but there have done little of work on the study and exploitation of active substance distributed in the skin of Chinese Forest Frog. There are many bioactive substances in the skin of frogs, which have different special molecule structures and complicated biological functions. These bioactive substances have important value in clinical application, and great potential for the exploitation and utilization of these resources. As much, there are lots of works will be done. It will be a firm foundation of more effective exploitation and utilization of Chinese Forest Frog resource, if some important value bioactive substances can be isolated and extracted from the skin of Chinese Forest Frog, and richen the "North medicine" resources.The experiment chose the skin of Chinese Forest Frog bought from the "Maoer mountain" of Hei Longjiang province as experimental material. The optimal extracted condition of active peptides and technical route was confirmed through conditional experiment of extracted system, extracted temperature, extracted hour. The data of SDS-PAGE electrophoresis, which analyzed small molecule peptides was confirmed through probing, and established the experiment project of isolation and purification of active peptides in frog skin by PAGE electrophoresis.Antibiotic test was chose to examine the functions of active peptides in the skin of Chinese Forest Frog. Bacterium: E. coli, Pseudomonas, Staphylococcus aureus, Actinomyces. The result suggests that the small molecule bioactive peptide in the skin of Forest Frog has ability against gram-positive bacterium and gram-negative bacterium. This indicates active peptides of Forest Frog skin have great potential for the exploitation and utilization.
