北洋金花的化学成分研究

Studies on the Chemical Constituents of Flower of Datura Innoxia Mill

【摘要】 北洋金花为茄科曼陀罗属植物毛曼陀罗Datura innoxia Mill.的干燥花。临床上常用其作为《中国药典》收载的正品洋金花[白曼陀罗(Datura metel L.)的花]的代用品应用。为了阐明两者化学成分的异同,本文对北洋金花进行了化学成分研究。 本文采用阳离子交换树脂、大孔吸附树脂柱色谱等方法精制、富集北洋金花的非生物碱水溶性组分,并应用硅胶柱色谱、聚酰胺柱色谱、ODS反相柱色谱等方法进一步进行分离。结果共分离得到8个单体化合物。结合理化分析和通过对其ESI-MS谱、1H-NMR谱、13C-NMR谱、DEPT谱的综合解析,鉴定其中6个单体化合物化学结构分别为7-O-α-L-鼠李吡喃糖基-山柰酚(7-O-α-L-ramnopyranosyl-Kaempferol)(Ⅰ);7-O-β-D-葡萄吡喃糖基-山柰酚(7-O-β-D-glucopyranosyl-Kaempferol)(Ⅱ);3-O-β-D-葡萄吡喃糖基、7-O-α-L-鼠李吡喃糖基-山柰酚(3-O-β-D-glucopyr anosyl,7-O-α-L-ramnopyranosyl-Kaempfer-ol)(Ⅲ);3-O-β-D-葡萄吡喃糖基(1→2)-β-D-葡萄吡喃糖基-山柰酚[3-O-β-D-gluco pyranosyl-(1→2)glucopyranosyl-Kaempferol](Ⅳ);3-O-β-D-葡萄吡喃糖基(1→2)-β-D-葡萄吡喃糖基、7-O-α-L-鼠李吡喃糖基-山柰酚[3-O-β-D-glucopyranosyl(1→2)-glucopyranosyl,7-O-α-L-ramnopyranosyl-Kaempferol](Ⅴ)和3-O-β-D-葡萄吡喃糖基(1→2)-β-D-葡萄吡喃糖基、7-O-β-D-葡萄吡喃糖基-山柰酚[3-O-β-D-glucopyranosyl(1→2)glucopyranosyl,7-O-β-D-glucop yranosyl-Kaempferol](Ⅵ)。这6个化合物均为首次从北洋金花中分离获得,同时证明,北洋金花的黄酮类的种类与洋金花基本相同。 本文确定结果可为阐明北洋金花和洋金花化学成分异同,进而深入比较两者功效的异同,以及开发利用北洋金花的资源提供了有价值的科学依据。

【Abstract】 Datura innoxia is dry flower of Datura innoxia Mill. It belongs to the family of Solanaceae. And it can be the substitute of Datura metel L.which is recorded in the <Chinese Pharmacopoeia>.To illustrate the difference of their Chemical constituents the article studied ones of Dutura innoxia Mill..By ion exchange resin 732(001x7) and AB-8 Macropore Adsorb ing resin the water-solubility component except alkaloid was refined and enriched.The further separation was made by using silicagel column chromatography, ODS column chromatography and polyamide chromatography.As a result,eight compounds (1-8) had been isolated from the 50% effective fraction of treatment psoriasis, using 1H-NMR,13C-NMR,compounds:7-O-a-L-rhamnopyranosyl-Ka empferol,7-O-β-D-glucopyranosyl-Kaempferol,3-O-β-D-glucopyran osyl,7-O-a-L-rh-amnopyranosyl-Kaempferol,3-O-β-D-glucopyranos yl-(1-2)-β-D-glu-copyranosyl-Kaempferol,3-O-β-D-glucopyranos-yl-(1-2)-β-D-glucopyranosyl,7-O-a-L-rhamnopyranosyl-Kaempfer ol and 3-O-β-D-glucop yranosyl (1-2)-p-D-glucopyranosyl,7-O-B -D-glucopyranosyl-Kaempferol.The compounds (1-6) were isolated from Datura innoxia Mill, for the first time.At thesame time,the sort of flavonoid in Datura innoxia Mill.and Datura metel L.is essentiall y the same.The result from this article can offer valuable scientific basis f or illuminating the similarities and differences between the chemica 1 constituants of Datura innoxia and Datura metel L., comparing thei r efficiency , and empoldering the resources of Datura innoxia.
