大豆黄酮对大鼠肌肉生长、内分泌水平与细胞免疫功能的影响

【摘要】 本文主要研究了大豆黄酮对雄性大鼠肌肉生长、内分泌水平及其细胞免疫机能的影响。 一、大豆黄酮对雄性大鼠增重和肌肉生长的影响 实验选择健康的初始体重在200g左右的5周龄SD雄性大白鼠,按年龄体重相近的原则随机分为三组。第一组每天每只灌服大豆黄酮50mg/kg,第二组每天每只灌服大豆黄酮100mg/kg,第三组每天每只灌服花生油做空白对照,持续四周。在实验结束时称重,分离腓肠肌、心肌并称重。在刚分离的腓肠肌和心肌上取大小为0.2×0.3m~2的组织块,做石蜡切片。显微镜下观察测量肌纤维密度、直径与脂肪细胞密度、直径。结果表明:50mg/kg大豆黄酮组大鼠与对照组大鼠相比,实验期內增重上升7.17%,,心肌重提高3%,腓肠肌重上升10.74%,腓肠肌体重比下降0.4%,差异均不显著;腓肠肌肌纤维直径大小提高14.49%(p<0.05),肌密度上升1.21%,脂肪细胞直径大小下降8.78%,脂肪细胞密度上升5.45%,差异均不显著;心肌纤维直径大小提高23.31%(p<0.01),心肌密度下降2.1%,心肌脂肪细胞直径大小下降8.91%,心肌脂肪细胞密度不变。100mg/kg大豆黄酮组大鼠与对照组大鼠相比,实验期内增重下降5.86%,心肌重下降8%,腓肠肌重下降3.01%,腓肠肌体重比上升0.05%;心肌纤维直径大小下降10.74%,肌密度上升2.43%,脂肪细胞直径大小下降4.88%,脂肪细胞密度上升3.64%;心肌纤维直径大小下降4.14%,心肌密度上升1.1%,心肌脂肪细胞直径大小下降6.5%,心脂肪细胞密度下降1.76%,差异均不显著。由实验可知50mg/kg剂量的大豆黄酮能显著提高心肌、腓肠肌的肌纤维直径,而100mg/kg剂量的大豆黄酮对肌纤维能使肌纤维直径下降,但差异不显著。 二、大豆黄酮对雄性大鼠内分泌水平的影响 将大鼠随机分为三组,每组7只大鼠,每组分别灌服大豆黄酮50mg/kg、大豆黄酮100mg/kg、花生油。在实验结束时,对大鼠眼窝静脉丛采血,颈椎脱臼处死。离心分离血清用放射免疫法测定其血清中激素含量。结果表明:与对照组相比,50mg/kg大豆黄酮组实验期内睾酮含量提高23.89%(p<0.05),GH上升15.07%(p<0.05)。与对照组相比,100mg/kg大豆黄酮组实验期内睾酮含量下降51.25%(p<0.01),GH下降13.38%(p<0.05)。由实验可知,50mg/kg剂量的大豆黄酮能提高睾酮、GH的含量,刘兆斌:大豆黄酮对雄性大鼠肌肉生长、内分泌水平与细胞免疫功能的影响差异显著;1 00mg/kg剂量的大豆黄酮能降低辛酮、GH的含量,差异显著。三、大豆黄酮对雄性大鼠细胞免疫功能的影响 在实验结束时,对大鼠眼窝采血,采集肝素钠抗凝血,计数RBC数和WBC数,酸性非特异性醋酶(ANAE)复合对比染色法测定外周血淋巴细胞.结果表明:与对照组相比,50mg/kg大豆黄酮组,RBC数上升33.19%(p<0.05),WBC数上升18.18%(p>.05),T淋巴细胞ANNE阳性率上升15.86(p<0.05)。与对照组相比,100mg/kg大豆黄酮组RBC数下降3.63%(p>.05),WBC数下降3.1%(p>0.05),T淋巴细胞ANNE阳性率下降6.3%(p>0.05). 由以上实验研究可知,50mg/kg大豆黄酮能显著提高大鼠的辛酮、生长激素含量;并能显著提高心肌和排肠肌肌纤维的直径;显著提高RBC数,T淋巴细胞阳性率。1 00mg/kg大豆黄酮能显著降低大鼠辛酮含量,生长激素水平也呈下降趋势,但变化不显著;能促使肌纤维直径变小;细胞免疫功能下降,但均不显著。可见在体内大豆黄酮对肌肉生长、内分泌水平呈现一定剂量依赖效应,但1 00mg/kg大豆黄酮对大鼠细胞免疫功能的抑制作用不明显.

【Abstract】 The effects of daidzein on muscle growth,the level of endocriones and cellular immune function in male rats were studied in this thesis.The presetent research includes three parts,and the result are shown as follows:1. Effects of Daidzein on Rats Growth and Muscle Growth in male RatsTwenty-four growing male rats were allotted randomely into three groups.The first growp(I group) was inspired daidzein of 50mg/kg body weight day for 30 days,the second group (II group) was inspired daidzein of 100mg/kg body weight dayfor 30 days,the control group(III group) was inspired peanut grease for 30 days.Compared to those of control group. The first growp increased body weight gain by 7.17%, gastrocsemins musle/body weight decreased by 0.4%, diameter of cell of muscle of gastrocsemins muscle increased 14.49%(p<0.05), density of cell of muscle increased 1.21%, diameter of lipocyte decreased 8.78%, density of lipocyte increased 5.45%, the differences were indistinctly; diameter of cell of cardial muscle increased 23.31% (p<0.01), density of cell of cardial muccule increased 2.1%, diameter of cardial lipocyte decreased 8.91%, density of cardial lipocyte have no change.The second growp decreased body weight gain by 5.86%, gastrocsemins muscle/body weight decreased by 0.05%, diameter of cell of musle of gastrocsemins musle decreased 10.74%, density of cell of musle increased 2.43%, diameter of lipocyte decreased 4.88%, density of lipocyte decreased 3.64%; diameter of cell of cardial muccule decreased4.14%, density of cell of cardial muscle increased 1.1%; diameter of lipocyte decreased 6.5%, density of lipocyte decreased 1.76%,the differrences were indistinctly.2. The Level of Endocrinous in Male RatsCompared to those of control group, The first growp’s blood concentration of testosterone and GH were elevated by 23.89% (p<0.05) and 15.07% (p<0.05) .The second growp’s blood concentration of testosterone and GH were decreased by 51.25% (p<0.01) and 13.38% (p<0.05) .3. Effects of Daidzein on Cellular Immune Function in Male RatsThe first group thymus gland/weight increased by 16.88% (p<0.01) ,the Red Blood Count(RBC) and the White Blood Count (WBC) and the trend of ANNE positive rate of lymphocytes increased by 33.19% (p<0.05), 18.18% (p>.05),and 15.86% (p<0.05)0 Compared to those of control group, The second gthymus gland/weight increased by 25.65% (pO.Ol) ,the Red Blood Count(RBC) and the White Blood Count (WBC) and the trend of ANNE positive rate of lymphocytes decreased by 3.63% (p>0.05) , 3.1 % (p>0.05) ,and6.3% (p>0.05)0 These experiment indicate that low dosage of daidzein can promote the growth of rats and cellular immune function by increase the levels of endocrines hormone,white high dosage of daidzein can inhibit it.In conclusion, 50mg/kg Daidzein can improve the level of endocrines ,growth of muscle ,Cellular Immune Function in Male Rats.But the 100mg/kg Daidzein have the reverse effect
