仔猪OB-Rb mRNA与LHRH在前脑内共存及orexin A的分布定位

【摘要】 1 Leptin长形受体mRNA与LHRH在猪前脑内共存 本实验用原位杂交和免疫组织化学相结合的方法,研究了5头三元杂交仔猪前脑内leptin长形受体(OB-Rb)mRNA和黄体生成素释放激素(LHRH)免疫反应阳性物质的共存关系。实验结果表明,OB-Rb mRNA和LHRH免疫反应阳性神经元分布于下丘脑、海马结构、大脑皮层和杏仁核内共存,即上述结构内一些表达leptin长形受体mRNA的神经元也含有LHRH免疫反应阳性物质。在下丘脑,双阳性神经元主要在室旁核、室周核、腹内侧核、外侧区、弓状核;在大脑皮层内,双阳性神经元主要位于Ⅲ—Ⅴ层;在海马结构内,双阳性神经元主要位于齿状回的多形层和颗粒层和海马的锥体细胞层。实验结果提示,leptin可能通过直接作用于下丘脑的LHRH神经元调节动物的生殖和内分泌活动。 2 OrexinA在猪中枢神经系统和外周器官的分布定位 本实验用免疫组织化学方法研究了5头苏钟猪脑和外周器官内orexin A的分布定位。研究结果表明,在猪下丘脑、睾丸、肾上腺、胰腺、小肠、甲状腺和胸腺等部位都有orexin A免疫阳性细胞分布,在心未见orexin A免疫阳性细胞分布。在脑内,orexin A免疫阳性神经元主要分布于下丘脑的视前内侧区、室周核、室旁核、视上核、背内侧核、穹隆周核、乳头体核、前区、外侧区和后区等部位,以下丘脑外侧区、乳头体核和视上核出现的免疫阳性神经元最多,以室周核的最少。在外周器官,orexin A免疫阳性细胞大量存在于睾丸间质、胰岛和小肠的粘膜固有层,而在肾上腺皮质、甲状腺和胸腺等部位仅有少量免疫阳性细胞分布。本文结果与在羊上获得的结果基本相似,但也有差异。这些结果的异同,既说明orexin在种系发生上的保守性,也反映存在种间差异。

【Abstract】 1 Colocalization of OB-Rb mRNA and LHRH in the forebrain of pigletsIn order to find the morphological evidence that leptin plays a role in control of reproductive endocrine in the level of the hypothalamus, coexistence of OB-Rb mRNA and LHRH-like immunoreactive (LHRH-1R) in neurons of the pig forebrain was studied by combination of in situ hybridization and immunohistochemistry methods. The results! shown that both OB-Rb mRNA and LHRH-IR containing neurons(dual labelled neurons) were seen in the hypothalamus, hippocampal formation, amygdala and cerebral cortex of the pig, and that some of neurons expressing OB-Rb mRNA also containing LHRH-IR. It meant coexistence of OB-Rb mRNA and LHRH-IR in these neurons of the pig forebrain. In the hypothalamus, dual labeled neurons were seen in the paraventricular nucleus, periventricular nucleus, ventromedial nucleus, lateral area and arcuate nucleus. In the hippocampal formation, dual labeled neurons were mainly found in the polymorphic layer and the granular layer of the dentate gyrus and in the pyramidal layer of the hippocampus. In the cerebral cortex , dual labeled neurons were mainly observed in the III- V layers. The results suggested that leptin probably regulate endocrine and reproduction of animal through directly imposing on the LHRH neurons expressing OB-Rb in the hypothalamus.2 Localization of orexin A in the central nervous system and periphery organs of pigletsThe localization of orexin A in the brain and some periphery organs of pig was studied by immunohistochemistry. The results shown that orexin-like immunoreactive (orexin-IR) cells were observed in the hypothalamus, testis, adrenal gland, pancreas, small intestine, thyroid gland and thymus. In the hypothalamus, orexin-IR neurons were present in the medial preoptic nucleus, periventricular nucleus, paraventricularnucleus, supraoptic nucleus, dorsomedial nucleus, perifornical nucleus, mammillary nucleus, anterior area, lateral area and posterior area, with majority in the lateral area, supraoptic nucleus and mammillary nucleus and with minority in periventricular nucleus. In the periphery organs, orexin-IR cells were widely found in the testis, pancreas, small intestine and rarely observed in the adrenal gland, thyroid gland and thymus. Our results were similar to those obtained from the sheep. But there were some differences. These similarities and differences meant the conservative of phylogeny and species difference.
