粟酒裂殖酵母产辅酶Q10及其动力学研究

【摘要】 微生物发酵法生产辅酶Q10是最有希望实现工业化的生产方法,论文选取粟酒裂殖酵母S.promb 2.1794作为出发菌株,经过诱变得到辅酶Q10的高产菌S.promb B2.1794,研究了该菌的最佳培养基和发酵条件,经过优化后辅酶Q10的产量由9.64 mg/L提高到26.85 mg/L。进一步研究了在最优条件下的发酵动力学,建立了其动力学模型,为辅酶Q10的工业化生产提供了一定的指导。 微生物发酵法的一个缺点是发酵产物复杂、产率较低,从而使辅酶Q10的提取纯化比较困难,且测定成本较高。本文比较了常用的几种不同提取方法,确定以碱皂化法提取辅酶Q10,并对碱皂化提取法进行了优化;建立了高效液相色谱法和紫外分光光度法测定辅酶Q10含量之间的相关性,从而简化为以紫外分光光度法测定辅酶Q10。 目前用于微生物发酵法中的辅酶Q10产生菌的产率都较低,因而单纯依靠寻找辅酶Q10的高产菌株,其提高辅酶Q10产量的能力是有限的。论文探讨了通过微生物转化法将茄呢醇转化为辅酶Q10的可能性。通过实验确定了微生物转化法中底物的投料方式,即50 mL发酵培养基中,在发酵进行到18 h时添加50 mg以1%吐温80乳化的茄呢醇、37.5 mg的对羟基苯甲酸,并补加0.2 g/dL MgSO4,这时微生物转化法测得胞内辅酶Q10产量为27.89 mg/L,而发酵液中辅酶Q10产量达到22.34 mg/L,总的辅酶Q10产量达到了50.23 mg/L左右。

【Abstract】 The production of CoQ10 by microbe fermentation is the most hopeful method to be industrialized. The Schizosaccharomyces promb 2.1794 was selected as the wild strain in the research, a highly yielding strain, S.promb B 2.1794, was gained by the inducement. Fermented by S,promb B 2.1794 with optimal culture medium and fermentation conditions, the yield of CoQ10 was increased from 9.64 mg/L to 26.85 mg/L. The fermentation kinetics of CoQio under the conditions above was investigated and the kinetic models, which could guide the industrialized production of CoQ10, were established.The complexity of the fermentation product and the low output rate made the extraction and purification of CoQio very difficult and the cost of mensuration was also increased. This paper compared several common extraction methods and the alkali saponification method was chosen eventually. For the UV method could be used in measuring the CoQ10 extracted from fermentation output, the pertinence between the HPLC method and UV method used in the mensuration of CoQ10 was analyzed.At present, the product rate of the strains used in the fermentation was relatively low. The paper discussed the possibility of microbe transformation method which use the precursor compound such as solanesol and para-hydroxybenzoate as the substrate to product the CoQ10. By experiment, the adding mode of the solanesol and para-hydroxybenzoate were determined. The production of CoQ10 by microbe transformation method reached to 50.23 mg/L including 27.89 mg/L extracted from inner cell and 22.34 mg/L gained in the fermentation solution.
