安非他酮、羟基安非他酮及其类似物的合成研究

【摘要】 安非他酮、羟基安非他酮是抗抑郁新药。本文以间氯苯丙酮、苯丙酮、对苄氧基苯丙酮和6-甲氧基萘丙酮为原料,经α-溴代、2-氨基-2-甲基-丙醇、N-甲基乙醇胺的胺化反应合成新型抗抑郁药安非他酮、羟基安非他酮及其类似物。 以间氯苯丙酮为起始原料,经CuX2的α-卤代合成2-卤-(3-氯苯基)-1-丙酮,2-卤-(3-氯苯基)-1-丙酮在NMP溶剂中于室温下与叔丁胺反应合成抗抑郁药安非他酮。用NMP作反应介质合成安非他酮的反应时间从报道(以乙腈、甲醇等作溶剂)的几十个小时、几天大大的缩短到1小时。以间氯苯丙酮为起始原料,在国内首次通过引入手性辅助剂(2S,3S)-酒石酸二甲酯不对称合成(+)-安非他酮。合成(+)-安非他酮的路线简单可行,并且符合光学纯药物给药的发展趋势,因此具有重要的实际意义。 以苯丙酮、对苄氧基苯丙酮和6-甲氧基萘丙酮为原料,经α-溴代合成中间体2-溴-苯基-1-丙酮、2-溴-(4-苄氧基苯基)-1-丙酮和2-溴-(6-甲氧基-2-萘基)-1-丙酮。在NMP中分别与叔丁胺反应,快速、高收率合成1-苯基-2-〔(1,1-二甲基乙基)氨基〕-1-丙酮、1-(4-苄氧基苯基)-2-〔(1,1-二甲基乙基)氨基〕-1-丙酮和1-(6-甲氧基-2-萘基)-2-〔(1,1-二甲基乙基)氨基〕-1-丙酮等α-叔丁胺基芳基丙酮类化合物。 在国内首次由间氯苯丙酮的溴化产物2-溴-(3-氯苯基)-1-丙酮与2-氨基-2-甲基丙醇在NMP中反应合成新型抗抑郁药羟基安非他酮。本合成路线反应时间短,反应收率高、操作方便。 首次在国内以2-溴-苯基-1-丙酮、2-溴-(4-苄氧基苯基)-1-丙酮和2-溴-(6-甲氧基-2-萘基)-1-丙酮等中间体与2-氨基-2-甲基-丙醇反应,高收率合成有潜在抗抑郁活性的羟基安非他酮类似物2-苯基-3,5,5-三甲基-2-吗啉醇、2-(4-苄氧基苯基)-3,5,5-三甲基-2-吗啉醇和2-(6-甲氧基-2-萘基)-3,5,5-三甲基-2-吗啉醇三种新化合物。 首次在国内以间氯苯丙酮、苯丙酮、对苄氧基苯丙酮和6-甲氧基萘丙酮的α-溴化产物中间体与N-甲基乙醇胺反应,高收率合成具有潜在抗抑郁活性的化合物2-(3-氯苯基)-3,4-二甲基-2-吗啉醇、2-苯基-3,4-二甲基-2-吗啉醇、2-(4-苄氧基苯基)-3,4-二甲基-2-吗啉醇和2-(6-甲氧基-2-萘基)-3,4-二甲基-2-吗啉醇四种新化合物。
【Abstract】 Bupropion and hydroxybupropion were novel antidepressant. Novel antidepressant, bupropion, hydroxybupropion and their analogs were synthesized via a -bromination ,aminization of 2-amino-2-methyl-propanol and N-methylethanolamine from 3-chloropropiophenone, propiophenone, (4-benzyloxy)-l-propiophenone and (6-methoxy-2-naphthyl)-1- propiophenone.Intermediate,2-bromo-(3-chlorophenyl)-1-propiophenone,2-chloro-(3-chlorophen yl) -1-propiophenone that was synthesized via a -halogenation with CuX2 reacted with tert-butylamine in NMP at ambience to give antidepressant ,bupropion .The reacting time was cut short to 1 h from hours,days in NMP solvent. (+)-Isomer of bupropion was first asymmetrically synthesized with chiral auxiliary (2S,3S)-(-)-dimethyl tartrate from readily available 3-chloropropiophenone in china.The route was simple , available and met the tendency of administer using optically pure isomer. So,it had significance of reality .2-Bromo-phenyl-1 -propiophenone,2-(4-benzyloxy)-1 -propiophenone and 2-(6-methoxy-2-naphthyl)-1-propiophenone respectively reacted with tert-butylamine in NMP to give l-phenyl-2-[(1,1-dimethylethyl)amino]-1-propanone,1-(4-benzyloxy)-2-[(1,1-dimethylethyl)amino]-1-propanoneand 1-(6-methoxy-2-naphthyl)-2-[(1,1-dimethylethyl) amino]-1-propanone in high yield.The novel antidepressant , hydroxybupropion was synthesized first in NMP via 2-bromo-(3-chlorophenyl)-1-propiophenone from 3-chloropropiophenone in china. The route had the advantage of short time,high-yield and easy-operation.The novel analogs of hydroxybupropion ,2-phenyl-3,5,5-trimethyl-2-morpholinol, 2-(4-benzyloxy)-3,5,5-trimethyl-2-morpholinol,and2-(6-methoxy-2-naphthyl)-3,5,5-trimethyl-2-morpholinol were first synthesized in china by 2-bromo-phenyl-1-propiophenone,2-bromo-(4-benzyloxy)-1-propiophenone and 2-bromo-(6-methoxy-2-naphthyl)-1- propiophenone reacting with 2-amino-2-methyl-propanol in high yield .2-Bromo-(3-chlorophenyl)-1-propiophenone,2-bromo-phenyl-1-propiophenone, 2-bromo-(4-benzyloxy)-l -propiophenone and 2-bromo-(6-methoxy-2-naphthyl)-1 -propiophenone reacted in NMP with N-methylethanolamine to give four novelcompounds in high yield:2-(3-chlorophenyl)-3,4-dimethyl-2-morpholinol,2-phenyl-3,4 dimethyl-2-morpholinol,2-(4-benzyloxy)-3,4-dimethyl-2-morpholinol,2-(6-methoxy-2-naphthyl)-3,4-dimethyl-2-morpholinol.
