NF基因表达产物在大鼠脑的分布

Distribution of Immunoreactive Structure of NF Gene Product in Rat Brain Studied by Immunohistochemistry

【摘要】 为了解NF1基因表达产物蛋白质神经纤维素及NF2基因表达产物蛋白质Merlin在大鼠脑中的分布,本实验利用兔抗Merlin抗体、兔抗神经纤维素血清及小鼠抗胶质纤维酸性蛋白单克隆抗体,分别通过免疫荧光组织化学法及免疫组织化学ABC法,观察了10只成年雄性SD大鼠脑中神经纤维素及Merlin的分布。其结果如下: (1)神经纤维素阳性免疫反应较强的脑区和核团有:大脑皮质、海马结构CA1-CA3区、海马齿状回、丘脑前核群、丘脑内侧核群、丘脑中线核群及下丘脑室周核、室旁核等;呈中等阳性免疫反应的有:杏仁核、下丘脑前区、下丘脑外侧区、下丘脑后区及小脑的蒲肯野氏细胞层、颗粒层等;阳性免疫反应较弱的脑区和核团有:尾壳核、屏状核、苍白球、疆内侧核、疆外侧核、外侧膝状体核、内侧膝状体核、隔外侧核、隔内侧核、三叉神经脊束核、蜗神经背侧核、蜗神经腹侧核、前庭上核、前庭内侧核、巨细胞网状核等。 (2)Merlin阳性免疫反应较强的脑区和核团有:大脑皮质、杏仁核、丘脑前核群、丘脑内侧核群、丘脑中线核群、脑干前庭内侧核以及小脑皮质颗粒层等;呈中等阳性免疫反应的有:海马结构、隔外侧核、隔内侧核、下丘脑室周核、室旁核、背内侧核、腹内侧核等;阳性免疫反应较弱的脑区和核团有:尾壳核、屏状核、苍白球、下丘脑外侧前核、下丘脑背侧核、三叉神经脊束核、前庭上核、前庭脊核、动眼神经核及巨细胞网状核等。 (3)神经纤维素和Merlin除在神经细胞胞浆中有表达外,在细胞核内也有分布,在部分细胞还能见到突起染色。在星形神经胶质细胞中未能观察到神经纤维素及Merlin的阳性免疫反应信号。 由此认为神经纤维素和Merlin在正常大鼠脑中有广泛的表达,但在表达强弱上,在不同脑区以及在神经细胞与神经胶质细胞之间存在着明显的不均匀性。本实验结果为进一步研究NF基因的功能提供了形态学基础。

【Abstract】 To provide more morphological data for studying function of neurofibromatosis (NF) gene in nervous system, the distribution of immunoreactive structure of two NF gene products, Neurofibromin (coded by NF1 gene) and Merlin (coded by NF2 gene), in the rat brain, were investigated in ten adult SD rats. Immunofluorescence and immunohistochemistry (ABC method) were employed separately to demonstrate Neurofibromin using rabbit anti-neurofibromin serum and Merlin using rabbit anti-Merlin serum.Result: (1) Intense neurofibromin labeling cells were found predominately in the cerebral cortex, the field CA1-CA3 of ammon’s horn, the dentate gyrus, the anterior nuclear group of thalamus, .the medial nuclear group of thalamus, the midline nuclear group of thalamus, the periventricular hypothalamic nucleus, and the paraventricular hypothalamic nucleus. Moderate neurof ibromin labeling cells were found in the amygdala, the anterior hypothalamic area, the lateral hypothalamic area, the posterior hypothalamic area, and the purkinje layer of cerebellum. Weakly labeling cells were also found in the caudate putamen, the claustrum, the globus pallidus, the medial habenular nucleus, the lateral habenular nucleus, the lateral geniculate nucleus, the medial geniculate nucleus, the lateral septal nucleus, the medial septal nucleus, the nucleus of the spinal tract of the trigeminal nerve, the dorsal cochlear nucleus, the ventral cochlear nucleus, the superior vestibularnucleus, the medial vestibular nucleus, and the gigantocellular reticular nucleus.(2) Intense merlin labeling cells were found in the cerebral cortex, amygdala, the anterior nuclear group of thalamus, the medial nuclear group of thalamus, the midline nuclear group of thalamus, the medial vestibular nucleus, and the granular layer of cerebellum cortex. Moderate labeling cells were found in the hippocampal formation, the lateral septal nucleus, the medial septal nucleus, the periventricular hypothalamic nucleus, the paraventricular hypothalamic nucleus, the dorsomedial hypothalamic nucleus, the ventromedial hypothalamic nucleus. Weakly labeling cells were found in the caudate putamen, the claustrum, globus pallidus, the lateroanterior hypothalamic nucleus, the dorsal hypothalamic nucleus, the nucleus of the spinal tract of the trigeminal nerve, the superior vestibular nucleus, the spinal vestibular nucleus, the oculomotor nuclei, and the gigantocellular reticular nucleus.(3)Both neurofibromin and merlin immunoreactivities were observed in the cytoplasm and nucleus of neurons, Whereas, neither neurofibromin nor merlin signal could be found in the neuroglial cell.It is concluded that there is a wide distribution of neurofibromin and merlin in the rat brain, with different intensity showing in different area of the brain and between the neurons and neuroglial cells. This study provided morphological base for further study of the function of NF gene.
