青光安颗粒对兔急性高眼压视神经轴突保护作用的实验研究

【摘要】 目的: 观察青光安颗粒对兔急性高眼压状态后视神经轴突及视网膜神经节细胞的保护作用。 方法: 将48只家兔随机分成青光安治疗组、模型对照组,每组24只。均用生理盐水灌注法随机取一眼造成急性高眼压模型,模型组另一眼则穿刺灌注不加压作为自身对照眼(空白对照组),连续灌胃14d。分别于造模后7、14d处死,检测视网膜中NO及谷氨酸的含量,并观察视网膜的病理组织学改变。结果: 7d后,各组视网膜组织中NO、谷氨酸含量比较差异有显著性(P<0.01),治疗组和模型组中NO及谷氨酸含量均高于空白组,差异具有显著性(P<0.01,P<0.05),但治疗组明显低于模型组,差异亦具有显著性(P<0.01)。14d后,治疗组中NO及谷氨酸含量均接近空白组,差异无显著性(P>0.05);但模型组仍明显高于空白组,差异具有显著性(P<0.05,P<0.01)。治疗组与模型组两组两周之间比较差异均有显著性(P<0.05)。电镜结果表明,与模型组对比治疗组轴突肿胀较轻,轴浆内线粒体清晰数目较多,视网膜神经节细胞损伤较轻。 结论: 青光安颗粒能改善视网膜微循环,促进轴浆传输阻滞的恢复,加速急性高眼压状态后视网膜组织中NO及谷氨酸的清除,对视神经轴突、视网膜神经节细胞有保护性作用。

【Abstract】 Objective: To observe the protective effects of QingGuangAn Granule(QGAG) on optic nerve axons and retinal ganglion cells in rabbit eyes with acute ocular hypertension. Method : 48 rabbits were divided into two groups ( 24 in each). The acute ocular hypertension(60mmHg) models were established by forcing perfusion of normal saline solution into the anterior chamber maintained for 2h in one eye, the other eye in experimental group which was maintained at basic intraocular pressure by the same method was regard as normal control. Each group was irrigated into the stomach for 14 days. The levels of the glutamic acid and NO in retina was measured on 7 and 14 days respectively , and the pathologic changes were observed. Result: After 7 days, the glutamic acid and NO levels have significantly difference between each group (P<0.01), and the treatment group and experimental group were significantly higher than those in normal control group(P<0.05,P<0.01). But the treatment group was significantly lower than those in experimental group(P<0.01). After 14 days, the glutamic acid, and NO levels in treatment group were near to the normal control group(P>0.05). However in the experimental group was significantly higher than those in normal control group (P<0.05,P<0.01). It has significantly different between two weeks in the treatment group and experimental group (P<0.05). Electron microscope result indicated that compared with experimental group optic nerve axons swelled relatively gentler, mitochondria in optic nerve axons was clear and more , and the degree of damaged retina ganglion cells in treatment group was lesser than that in experimental group. Conclusion: QGAG can ameliorate retina microcirculation, promote the axoplasmic<WP=6>transport of optic nerve , accelerate NO and glutaminic acid remove in rabbits retina with acute ocular hypertension. It has the protective function for the optic nerve axons and the retinal ganglion cells.
