苦丁茶总萜类降脂作用的实验研究

【摘要】 目的:研究冬青科冬青属植物苦丁茶(Ilex kudingcha C.J.Tseng)中总萜类(total terpene)成分的降脂作用.方法:①SD大鼠喂饲高脂饲料,同时灌胃给予苦丁茶总萜类(kudingcha total terpene,KDC-TT),连续21天,观察对血清中TG、TC、LDL-C、HDL-C、脂蛋白a[Lp(a)]、载脂蛋白AI(ApoAI)、载脂蛋白B(ApoB)、脂质过氧化物(lipid peroxide,LPO)和肝脏、主动脉壁TG、TC的影响;②SD大鼠连续喂饲高脂饲料35天,从第22天开始灌胃给予KDC-TT 治疗14天,观察对血清TG、TC、LDL-C、HDL-C的影响;③NIH小鼠喂饲果糖造成急性高甘油三酯模型,观察KDC-TT对血清TG的影响。结果:KDC-TT预防给药可显著降低食饵性高脂血症大鼠血清中TG、TC、LDL-C及ApoB水平,降低动脉粥样硬化指数(AI,即TC/HDL-C)。同时可降低肝脏及主动脉壁中TG、TC水平。对HDL、ApoAI无明显影响。对血清过氧化脂质有一定的降低作用,在300mg.kg-1剂量可升高血清SOD及GSH水平。采用治疗给药方式,发现KDC-TT各剂量组仍可降低血清TG水平,高剂量组可降低血清TC及LDL-C水平。对急性高甘油三酯模型小鼠可明显降低血清TG水平。结论:KDC—TT能降低食饵性高脂血症大鼠血清TG、TC、LDL-C、过氧化脂质、ApoB水平,亦可降低肝脏和主动脉壁TG、TC水平,并可降低AI。能降低果糖所致急性高甘油三酯小鼠血清TG水平。
【Abstract】 Objective: To study the lipid-lowering effects of KDC-TT .Methods: We fed SD rats with rich-oil food for 21 days successively, meanwhile treating the animals by oral injection with KDC-TT,measured TC,TG,LDL-C ,HDL-C ,ApoAI , ApoB and LPO in the serum as well as TG and TC in the arterial wall and in the liver tissue. In another experiment, SD rats were fed with rich-oil food for 35 days, from the 22th day the animals were treated with KDC-TT by oral injection. TG,TC,LDL-C and HDL-C in the serum were measured to study the effects of the drug. At the same time, we fed the NIH mice with D-fructose to build acute hypertriglyceridemia model for studying the effect on decreasing TG in the serum.Results: KDC-TT significantly decreased the TC,TG and LDL-C levels in the serum of rats with hyperlipidemia . TG ,TC levels in the arterial wall and in the hepatic tissue declined. Atherosclerosis Index(AI) also declined. The concentration of Apo B decreased while HDL-C and ApoAI didn’t change<WP=5>significantly. LPO in the plasma was inhibited by KDC-TT , SOD and GSH raised in the group with a dose of 300mg.kg-1. Treating the hyperlipidemia with KDC-TT after the rats were fed for 21 days with a rich-oil food, we observed that the level of TG in the serum fall to a lower one , the same with the levels of TC,LDL-C in the highest dose group. For the mice with an acute hypertriglyceridemia,KDC-TT also showed a strong effect on decreasing TG level in the serum.Conclusion: KDC-TT possesses the effects of decreasing TG、TC、LDL-C、lipid peroxide and ApoB in the serum , decreasing TG and TC in the liver and in the arterial wall of rats with hyperlipidemia induced by feeding rich-oil food ; it also has the effects of reducing TG levels in the serum of mice with acute hypertriglyceridemia.
