不同采收期规范化栽培前胡总香豆素含量动态变化研究
Study on Contents of the Total Coumarines of Standard Cultivated Qian-Hu (Peucedanum Praeruptorum Dunn. and Peucedanum Decursirum Maxim.) in Different Collecting Times

【摘要】 前胡为常用中药材,中华人民共和国2000版药典一部前胡为伞形科植物白花前胡(Peucedanum praeruptorum Dunn.)和紫花前胡(Peucedanum decursirum Maxim.)的干燥根及根茎。20世纪60年代以来人们对前胡的化学成分尤其是香豆素类成份进行了较系统的研究,但尚未有人对前胡的最佳采收期进行科学的研究。通过对规范化栽培前胡不同采收期总香豆素含量变化进行研究,主要结论如下:1、采用了紫外分光度法测定前胡总香豆素,该方法的稳定性、精密度、重现性和回收率实验结果均良好。用此方法考查了不同采收期规范化栽培白花前胡与紫花前胡中总香豆素含量的变化情况,经过统计分析,同时结合原植物的生长情况,提出前胡合理的采收期应在11月中下旬。2、初步建立了栽培白花前胡与紫花前胡药材的高效液相指纹图谱方法,该方法的稳定性、精密度、重现性实验结果均良好。用此方法分别对栽培白花前胡与紫花前胡药材的高效液相指纹图谱共有峰的总峰面积进行考查,发现不同采收期总峰面积的变化与总香豆素含量动态变化相似,因此可作为一种质量控制与药材鉴别的辅助方法。 3、比较了7个地区收集的9个前胡样本总香豆素的含量及其指纹图谱。发现无论从总香豆素的含量还是指纹图谱峰的组成方面进行比较,都有较大的不同。由此可见市售前胡药材有效成分的种类与含量与药材是否按照GAP种植有必然的联系,同时佐证了规范化栽培中药材(GAP)在我国推行的重要性。
【Abstract】 Qian-Hu is a kind of common Chinese herb. There are two species in CP-2000:Peucedanum praeruptorum Dunn. and Peucedanum decursirum Maxim. Their part is the root and main biological active compeonents are coumarines. Since 1960s , there have been more reports on chemical constituents and pharmacology of P. praeruptorum Dunn. and P. decursirum Maxim., but no study on their collecting time. In order to determine the best collecting time of standard cultivated Qian-Hu, we made the study : 1. We used an ultraviolet-visible spectrophotometry(UV) assay method to determine the content of total coumarines in Qian-Hu. The recovery was 99.16% with RSD less than 1.55%. The method was simple,accurate and reproducible. We determined the contents of the total coumarines of standard cultivated P.<WP=5>praeruptorum Dunn. and P. decursirum Maxim. in different collecting times by the method. The contents of the total coumarines increased with growing of the plant. According to the results of statistics, and growing period of P.praeruptorum Dunn. and P. decursirum Maxim.,we concluded that the best collecting time of the standard cultivated P. praeruptorum Dunn. and P. decursirum Maxim. was November.2.To study the determination method of the fingerprint of Qian-Hu and to establish high performance liquid chromatography (HPLC) assay methods separately to determine the fingerprint of the standard cultivated P. praeruptorum Dunn. and P. decursirum Maxim. The results of stability experiment, accuracy experiment and reproducible experiment were accurated. We separately determined the area of total common peaks in HPLC fingerprint from different gathering time of the standard cultivated P. praeruptorum Dunn. and P. decursirum Maxim. The relative retention time and the ranges of relative area of the common peaks were determined. The results showed that the regularity of change of the area of total common peaks was similar to that of the contents of total coumarines of the standard cultivated P. praeruptorum Dunn. and P. decursirum Maxim.in different collecting time.Because the methods were selective and reproducible, the fingerprint could be used for the quality control and indentification of the standard cultivated P. praeruptorum Dunn. and P. decursirum Maxim.<WP=6>3. We also determined the samples from different collecting areas. From the HPLC fingerprints of nine Qian-Hu samples from seven regions,we found that there was a difference in the chromatography peaks.The contents of the total coumarines of the standard cultivated Peucedanum praeruptorum Dunn. and Peucedanum decursirum Maxim.were much higher than those of the other Qian-Hu samples. The contents of the total coumarines of the P. decursirum Maxim.were much higher than those of P. praeruptorum Dunn. We could conclude that the primary plant of Qian-Hu for sale and collecting time were confused .So we might carry out GAP.
