甘草多糖提取纯化工艺研究

【摘要】 豆科植物甘草(Glycyrrhiza uralensis Fisch.)的根及根茎是最常用、最重要的中药。甘草含有三萜类、黄酮类及多糖类等化合物,其中甘草多糖具有广泛的生物活性,具很大的开发潜力,有望成为一类新药。 以往只注重从甘草中提取甘草酸和甘草黄酮,而忽略了多糖的重要活性,将含有大量多糖类物质的固形物作为废弃物扔掉,造成浪费。针对这种情况,本论文对不同原料(甘草超临界萃取物、提甘草黄酮后固形物、提甘草黄酮和甘草酸后的固形物、提甘草黄酮后固形物的超声提取物、甘草根茎原粉)的多糖含量进行了比较研究,并以超临界提取过甘草黄酮后的剩余固形物为原料对甘草多糖提取纯化工艺进行了深入而系统的研究,确定了一套新的甘草多糖提取纯化工艺。 工艺条件如下: 1.甘草多糖提取 以水为提取溶剂,在80℃条件下,提取3小时,反复三次,每次水的用量为30ml/g原料。甘草多糖纯度为40.00%,收率为5.40%。 2.甘草多糖纯化 粗多糖采用大孔树脂去色素,Sevag法去蛋白,无离子水透析去小分子杂质,并用二乙基氨基纤维素(DEAE-32)、葡聚糖凝胶(Sephadex G-75)柱层析技术对甘草多糖进行了进一步纯化。多糖纯度达到96.15%,收率为0.57%。 本工艺充分利用了甘草资源,以超临界二氧化碳萃取甘草黄酮后的固形物为原料,有效地提取、纯化了其中的甘草多糖,为研制甘草多糖类药品提供了理论和实践基础。
【Abstract】 The radix and rhizoma of licorice are the most common and important Chinese traditional medicine. Licorice contains the compounds such as triterpenoids, flavonoids and polysaccharides and so on. Glycyrrhiza polysaccharides(GPS) have the extensive biology activity and the great exploitative potential. It can be expected to be the new medicine.Only flavonoids and glycyrrhiza acid was paid attention to be extracted from licorice ago, but the important activity of GPS was neglected . It caused waste because GPS was threw away as residue . Considering it, researches had been done to the extraction of GPS from different materials (extraction extractant of supercritical CO2, residue of extracting flavonoids, residue of extracting flavonoids and glycyrrhiza acid, ultrasonic extractant of residue of extracting flavonoids,radix and rhizoma of licorice) and deeply and systematically researches had been done to the extraction and purification of GPS from the residue of extracting flavonoids. The new process for extraction and purification is decided.The condition of the process as follows:1. Extraction of GPS Use water to dip raw material for 3 hours, the volume of the water was 30ml/g raw material for one time, the temperature 80℃, repeated 3 times. The content of GPS is 40.00% and the yield is 5.40%.2. Purification of GPS The large-diameter adsorbent resin removing pigment, the method of Sevag removing protein, deionized water dialysis removing the impurity of small molecule was used in the purification of GPS and the further purification was finished using the column chromatography of DEAE-32 and Sephadex G-75. The content of GPS is 96.15% and the yield is 0.57%.This process used the licorice resource sufficiently, extracted and purified GPS from residue of supercritical CO2 extracting flavonoids effectively, and provided the strong base of theory and practice to develop polysaccharides medicine.
