医药中间体β-对羟基苯乙醇合成与工艺研究

【摘要】 研究了医药中间体β-对羟基苯乙醇的合成方法,对它的两条不同的合成路线进行了比较。对所合成的β-对羟基苯乙醇以及各种中间产物,利用红外光谱、核磁共振等进行了结构表征,利用熔点、高效液相色谱进行了纯度分析。讨论了反应时间、反应温度、反应物物质的量的比等条件对各化合物合成的影响,对反应机理进行了初步探讨,建立了β-对羟基苯乙醇的高效液相色谱分析方法。 在苯乙醇合成法中,以苯乙醇为原料通过酯化、硝化、脱酯、还原、重氮化这五步反应合成了β-对羟基苯乙醇,总收率达到24.0%。利用沸石分子筛进行选择性硝化的研究,使产物的o/p比为0.267。 在酯还原合成法中,由对羟基苯乙酸经酯化、还原合成了β-对羟基苯乙醇,两步反应的总收率为51.2%。利用正交实验得到了反应的最佳工艺条件。

【Abstract】 The two synthetic methods of -(4-hydroxyphenylethyl) alcohol is discussed in this paper. The different of them is proposed also. The product and its intermediates were confirmed by IR/H NMR and the purity of them was analysed by Melting Point and HPLC. The effects of various conditions to reaction were studied. The reaction mechanisms were discussed.In phenylethyl alcohol method -(4-hydroxyphenylethyl) alcohol was synthesized by esterification,nitrofication,reduction and diazo-tization from -phenylethyl alcohol in 24.0% overall yield. The number of o/p is 0.267 by selective nitrofication because of molecular sieve.In ester reduction method the 51.2% yield of product was obtained from 4-hydroxyphenylacetic acid via two steps. The optimum conditions were attained by the orthogonal experiment.
