卵泡发育及排卵障碍的肾虚血瘀动物模型研究

【摘要】 目的:寻找有助于评价资冲颗粒补肾化瘀促卵泡发育及排卵药效的动物模型。方法:对羟基脲所致肾虚模型、氢化可的松加肾上腺素所致血瘀模型、单一孕激素避孕药DMPA、LNG、癸息汤所致排卵抑制模型的血液流变学、血清E2、P、卵巢、子宫指数、卵巢生长卵泡、成熟卵泡、闭锁卵泡、黄体数、子宫内膜厚度、腺体数等指标进行考察。结果:羟基脲所致肾虚动物模型卵巢指数降低,卵泡数减少,血液流变学异常低下。氢化可的松加肾上腺素所致血瘀动物模型出现血粘滞度增高,但未出现明显卵泡发育及排卵抑制。孕激素及癸息汤所致的动物模型出现不同程度的血粘滞度升高,生长卵泡、闭锁卵泡数增多,黄体数明显减少,内膜变薄,子宫腺体数量少,孕激素水平低下。其中,血液流变学的改变以癸息汤组明显,排卵抑制以孕激素组较为明显。结论:孕激素所致的排卵抑制的血瘀动物模型可进行资冲颗粒补肾活血促排卵的药效学研究,癸息汤的排卵抑制作用尚需进一步研究。
【Abstract】 OBJECTIVE To find an animal model that helps to evaluate the effect of Zichong particles whose function is to tonify kidney, dissolve blood stasis, promote follicular development and ovulation. METHLDS: To inspect the markers such as hemorheology, estradiol, progesterone, the exponent of ovary and uterus, the number of growing follicle,ripe follicle, atresic follicle and luteum, the depth of endometrium, the number of glands of kidney deficiency model results from hydroxyurea, blood stasis model results from cortisol and epinephrine . the blockade of ovulation animal model with kidney deficiency and blood stasis results from sigle progesterone contraceptive DMPA,LNG and Traditional Chinese medicine. Results: The ovary exponent and the number of follicle of the kidney deficiency model results from hydroxyurea reduced, but the model came out without blood stasis; The blood stasis model results from cortisol and epinephrine had abnormal hemorheology, but without blockade of follicular development and ovulation; The models result from progesterone contraceptive and Traditional Chinese medicine had different degrees of abnormal hemorheology , increasing growing follicle and atresic follicle, reduced luteum and glands, low progesterone level, which imply that progesterone has the function of making the rats’ hemorheology abnormal and ovulation inhibition. Conclusion: The ovulation inhibition and blood stasis animal model results from progesterone can help to evaluate the effect of Zichong particle and other Traditional Chinese medicine which can tonify kidney, dissolve blood stasis and promote ovulation.
